With each addition of protein there is a reaction between the metal M and protein P to form M_aP_b according to aM+bP(?)M_aP_b The concentration of metal not bound to P, designated as [M], which is in equilibrium with M-aP_b is determined by ASV after each addition and gives an ASV response i_s=k[M], where i_s is the peak current of the stripping curve, and k is a proportionality constant. The titration curve has two regions. There is less metal reacted with P in the upper region so the ASV response is high. After the equiyalent point exists the lower region where most of the metal has reacted with P. The equivalent point (E_p) of the titration is the point at which theocncentration of added metal, C_M,is equal to (a/b).C_P, where C_P is the analytical concentration of protein. The following relationship can be obtained For the case of a 2:1 complex, for instance,a plot of i_s vs·[1/2C_M/(C_P-1/2C_M)]~(1/2) has a slope k/K~(1/2). Therefore it is posible to estimate the composition of the leadprotein complex and K. From the E_P of the titration ourve the MW of protein can be calculated. The following results were found: lmol of serum protein reacted with 2 mols of lead to form complex M_2P. The K'_s of lead-human albumin, learl-bovine albumin and lead-human gamma globulin are 1.3×10~6(pH=6), 9.4x10~5(pH=7.4) and 1.82×10~5(pH=7) respectively. The MW of human albumin, bovine albumin and human gamma globulin are 69650, 80000 and 221000 respectively, the results agree with that obtained by the classical method reported in the literature.

Phosphorylated chromosomal non-histone proteins (P-NHCP)of Hep. A cells and normal mouse liver cells have been studied. The contents of chromosomal non-histone proteins (NHCP) and P-NHCP in Hep. A cells are respectively 2.2 and 4.4 times those in normal liver cells. The ratio of P-NHCP to NHCP is also 2 times that in normal cells. The phosphate content of NHCP in Hep. A cells also increases, being 1.36 times that in normal cells. But the phosphate group per unit weight of NHCP and P-NHCP decreases to 65% and 32% of normal cells respectively. The SDS-gel slab electrophoresis pattern of P-NHCP in Hep. A cells demonstrates that a band with a molecular weight of about 69, 000 daltons is evidently deeper when compared with the normal one, and an additional band with a molecular weight of 47,000 daltons appears. The lower pI fractions of P-NHCP increase in isofocusing electrophoresis patterns of Hep. A cells. The amino acid composition of P-NHCP shows that the amount of accepters of the phosphate group is almost the same. The experiment indicated that the P-NHCP changes quantitatively and qualitatively in Hep. A cells.
