Objective To establish osteoblasts osteoclasts co culture system in vitro for the study of the biological characteristics of osteoblasts and osteoclasts and also to analyze the interactions between osteoblasts and osteoclasts. Methods The osteoblasts and osteoclasts were isolated from cancellous bone in illium. Osteoblasts osteoclasts co culture system was built to prevent these two kinds of cells from contact with each other although with the facility of their medium exchange. The osteogenety of osteoblasts was evaluated by MTT and ALP activity; the resorption activity of osteoclasts was investigated by TRAP activity and the area of absorption lacuna. Results Osteoblasts are full fusiform, ALP stain positive cells, while osteoclasts are multinuclear and TRAP stain positive, with the ability of forming bone lacuna; while in the co culture system, the proliferation of osteoblasts in co culture (0.60±0.08)OD is quicker(P=0.000)than that in mono culture (0.36±0.03)OD, and the ALP activity increased significantly (P=0.000)from (18.33±0.34)u/mg to (23.37±2.48) u/mg; while the average area of bone resorption formed by osteoclasts [(6.55±0.34)×10-2] μm2 was also increased obviously(P=0.000)from [(5.15±0.17)×10-2] μm2. Conclusion In co culture system, osteoclasts were able to enhance ossification effects of osteoblasts by accelerating their proliferation and differentiation; at the same time, osteoblasts upregulated the ability of bone resorption of osteoclasts. This implied that osteoblasts and osteoclasts can promote each others function. This study provide reliable model for bone metabolism research.

Conclusion:Puerarin can suppress the bone resorption and promote the bone formation, but the former effect is stronger than the latter.

Objective To observe the effect of the "Bushen" Chinese Medicine on IL-6, IL-6R and gp130 gene expression in bone marrow microenvironment and the bone marrow derived osteoclast formation and function. Methods Healthy 3 month old SD rats were randomly divided into 4 groups: sham-operation control group(S) , ovariectomized group(O), estrogen treated group(E) and Chinese medicine treated group(Z) . The femur bone marrow cells were collected and cultured at the 6th and 12th week after treatment. The number of the osteoclasts were measured with positive TRAP stain and IL-6,IL-6R,gp130 gene expression of bone marrow cells were measured by RT-PCR. Results The number of the bone marrow derived osteoclasts in group O were significantly increased compared with group S in either 6th or 12th week. Estrogen and Chinese medicine treatment both significantly inhibited the osteoclast formation and the effect was a little stronger in group E. The mRNA expression of IL-6 and IL-6R in group O were both higher than group S in the 6th week and declined afterwards in 12th week. It was demonstrated that the estrogen and Chinese medicine treatment could inhibit IL-6 and IL-6R mRNA expression both in 6th and 12th week. There was not significantly change in gp130 mRNA expression. Conclusions The results suggest that increased formation of bone marrow derived osteoclast is an important cause of bone resorption in ovariectomized rats. And this effect is closely correlated with the increased expression of IL-6 and IL-6R gene. The "Bushen" Chinese medicine can partly inhibit the bone marrow derived osteoclast formation and IL-6, IL-6R gene expression in bone marrow microenvironment, but no effect is observed on gp130.
