Acute toxicity (15min-EC 50) determination of 16 substituted naphthalene compounds to Photobacterium phosphoreum was undertaken according to the standard procedures, while the effects of molecular structures of selected compounds on their toxicity to test microorganism were logistically conducted by using quantitative structure-activity relationship (QSAR) technique. The relationship was developed as: -logEC 50 =5.592-7.489qH ++0.777 E LUMO + 0.009α ( n =16, r 2 adj= 0.970, S = 0.089, p <0.0001). The prediction model was tested with several different methods and the results indicated that the model was reasonable and robust. Furthermore, the quantum chemical parameters entering into QSAR model were used to discuss the possible toxicity pathways, and the results showed that the selected compounds were of reactivity and their toxicity behaviors were complex processes containing physical partition stages as well as bio-chemical reaction stages.

By substituting assembled chimaeras for corresponding region of pETPPGA, different recombinants were constructed and expressed in E. coli JM109(DE3). Mutants with obvious β-lactam synthetic activity were selected from the plates and the ratios of synthesis to hydrolysis (S/H) were determined subsequently. It was shown that the primary structures of selected positives exhibited significant diversity among each library. The best mutant possessed 40% higher synthetic activity than the wild type enzyme of PrPGA. It was further proved in this study that the domain of α subunit contributed much more to improve the specific activity of synthesis. Results showed a recombinant PGA with higher synthetic activity was acquired by the method of DNA shuffling.
