In order to investigate the pathogenetic mechanism of endotoxin shock at the cellular membrane Ievel and the anlishock mechanism of anisodamine, the effect of E. coli endotoxin and anisodamine on Ca2+-induced aggregation of artificial biological membrane, liposomes, was observed. The results showed that neither endotoxin alone, nor anisodamine alone induced aggregation of PC/PA/cholesterol liposomes. Endotoxin, however, preincubated with liposomes, significantly prompted Ca2+-indvced aggregation...

Endotoxin, however, preincubated with liposomes, significantly prompted Ca2+-indvced aggregation of liposomes. This effect of endotoxin is time-and dosedependent. Anisodamine, when preincubated with liposomes, did not alter Ca2+-induced aggregation of liposomes, though it significantly inhibited the endotoxn-prompted effect on the Ca2+-induced aggregation of liposomes. The effect of anisodamine on the biological membrane could explain, at least partly, the antishock mechanism of anisodamine against septic shock.

内毒素或山莨菪碱单独使用均不引起磷脂酰胆碱—磷脂酸—胆固醇脂质体的聚集反应。但内毒素与脂质体预先孵化,明显促进Ca~(2+)引起的脂质体聚集反应,其效应呈剂量依赖关系。山莨菪碱与脂质体预先孵化,对Ca~(2+)引起的脂质体聚集反应无明显影响,但显著抑制内毒素对Ca~(2+)引起的脂质体聚集反应的促进作用。山莨菪碱对内毒素的抑制效应呈剂量依赖关系。
