目的:建立杏仁核点燃的大鼠慢性癫痫模型。方法:在3.6%水合氯醛麻醉下,用漆包镍镙双极螺旋电极植入Wistar大鼠杏仁核,术后第7天,用微电极刺激大鼠杏仁核,记录脑电活动,同时观察大鼠行为变化。当脑电图上有发作后电位,即2~5Hz爆发性的尖波或棘慢波,波幅大于200μV,持续3s以上,同时按Racine分级法大鼠的行为表现达到4级或5级时可视为大鼠癫痫模型点燃成功。结果:34只Wistar大鼠被点燃。癫痫发作模式具有规律性、容易辨认、可重复诱发,癫痫发作程度可以量化的优点。6只为亚临床发作(即有典型的脑电图的棘波放电表现,但是行为不足4~5级)。结论:通过微电极刺激Wistar大鼠杏仁核可以建立稳定的大鼠癫痫慢性点燃模型。

Aim: Investigating to establish a chronic kindling epilepsy animal model of anti-brain antibody.Methods:The hippocampus CA1 area of Spreague-Dawley (SD) rat was continuously injected with 1 μl presynaptic membrane protein(Munc 18) anti-brain antibody. Injection was performed every other day, which altered every three weeks after fifth times. Injection finally lasted for ten weeks. During injection period, both eletroencepalograph(EEG)and epileptic behaviors of rat were monitored. After three months, rats were executed, whose brain slices were stained by HE and nissle methods. Results: Ten of twelve rats(83%) had epileptic EEG,five cases(50%) among who retained it after sixth week.Eepileptic behaviors from the first to the fifth grade were found in nine rats (75%), four cases (44%) among who retained that after sixth week. Under microscope, serious lesion, proliferating glial cell, reduced numbers and morphologic abnormality of neuron cell from stainde brain slices were found. On the other hand,only two rats with epileptic EEG and one with epileptic behavior were respecticely observed in control groups( P < 0.01) . Conclusion : Munc 18 antibody could induce epilepsy by chronic kindling way. However, its mechanism correlating epilepsy need to further investigate.

目的:探索建立一种慢性点燃抗脑抗体癫癎动物模型。方法:在SD大鼠的海马CA1区间断持续注射抗脑抗体——神经突触前膜蛋白(Munc-18抗体)1μL,隔天注射一次,5次之后每隔2周注射一次,共10周。监测注射前后大鼠的脑电图(EEG)和癎样行为。3个月后取脑切片,HE和尼氏染色后镜下观察。结果:实验组12只大鼠中有10只有癎样EEG(83%),6周之后有5只(50%)保留;9只有1~5级癎样行为(75%),第6周后保留4只(44%);而所有对照组中,仅2只大鼠有轻度异常脑波和1只有2级癎样行为,相比有显著差异(P<0.01)。镜下发现实验组损伤灶明显,胶质细胞增生,神经元细胞减少且形态异常。结论:Munc-18抗体能慢性点燃致癎大鼠,但其作用机制需进一步研究。

Results Ten of 12 rats (83%)produced epileptiform EEG, 5 cases (50%) among which maintained abnormal EEG after 6 weeks. Nine of the above 12 rats(75%)showed epileptiform behaviors of Racine grade 1 to 4, 4 cases (44%) among which had that after 6 weeks. In hippocampus, neurons showed less numbers and morphologic abnormality, while glial cells proliferated. TUNEL-positive cells significantly increased. Conclusion Munc-18 antibody could induce epilepsy by chronic kindling way which mechanism might be linked to apoptosis in area of hippocampus .However, it needs to be further investigated.

目的 研究抗脑抗体———神经突触前膜胞内蛋白 (Munc 18)抗体是否能慢性点燃致痫大鼠以及其致痫的机制。方法  在SD大鼠的海马CA1区注射Munc 18抗体 ,隔天注射一次 ,5次之后每隔两周注射一次 ,共 10周。期间监测大鼠的脑电图 (EEG)和痫样行为。 10周后取脑切片作HE、尼氏和TUNEL染色。 结果  实验组12只大鼠中 ,有 10只有痫样EEG(83% ) ,5只 (5 0 % )在 6周之后仍有保留 ;9只有 14级痫样行为 (75 % ) ,6周以后为 4例 (4 4% ) ;染色发现在海马区有胶质细胞增生 ,神经元细胞数量减少且形态异常 ,凋亡细胞数量明显增多。与对照组相比均有显著差异 (P <0 .0 1或P <0 .0 5 )。 结论  Munc 18抗体能慢性点燃致痫大鼠 ,其机制可能与海马区神经元细胞的凋亡有关 ,但还需进一步的研究证实。
