本实验使用人胎成骨细胞(OB)培养物做为氟中毒模型。通过形态学观察及细胞蛋白含量、碱磷酶活性、细胞放射性钙摄入率等生化指标测定,分析了氟的细胞毒作用。证实了氟中毒情况下氟离子可以影响 OB 的多种代谢途径;氟抑制 OB 生长、功能的浓度不在同一水平,不同的氟中毒浓度对 OB 功能产生双相作用。

The effect of 3,5,3′-triiodothyronine (T_3) on the differentiation and proliferationof osteoblasts (human fetal bone-cells) cultured in serum-free medium was investi-gated by assays of alkaline phosphatase(ALP) activity, intracellular cyclic AMPlevel and collagen synthesis in the cell. The result suggested that T_3 in physiologicconcentration stimulated the differentiation of osteoblasts in vitro, and might beinvolved in bone formation in vivo.

本文利用无血清培养的人胎成骨细胞(OB)体外模型,通过对 OB 内碱性磷酸酶(ALP)活性、胶原合成、DNA、蛋白含量及 cAMP 水平的测定,研究了生理浓度的 T_3(3,5,3′-三碘甲腺原氨酸)对 OB 生长、功能、分化的作用,结果显示:生理浓度的 T_3可显著地增加 OB内 ALP 的活性,并有时间相关关系,T_3作用96小时可显著地增加~3H-脯氨酸掺入到胶原蛋白中;然而,在96小时时期内,却未显著地改变细胞内蛋白及 DNA 的含量。这些结果表明.T_3引起 ALP活性、胶原合成的增加,不是由于促进了 OB 的增生,而是直接促进了体外培养的 OB 的分化而增加其功能。本研究提示,T_3在生理浓度下与体内骨形成有关。

Effects of copper vapour laser in enhancing the proliferative activity of hematopoieticGM-CFUc were studied with the bone marrow from new born mice and buman fetus. After theirradiation of hematopoietic stem/progenitor cells of mice by copper vapour laser of both578.2 and 510.6nm and cultured in vitro, the GM-CFUc increased significantly than thatof the nonirradiated control group. The enhancing effect of copper vapour laser to the pro-liferative human fetal bone marrow cells was similar to that of fetal liver hematopoieticcells treated by 514.5nm Ar~+ laser previously presented elsewhere.

本研究用铜蒸气激光照射人胎骨髓或小鼠骨髓造血干/组细胞,可以使造血细胞GM-CFUc增殖,用578.2nm和510.6nm的激光照射造血细胞,经体外培养后,其GM-CFUc值和未照射组比较都有显著增加,这些结果与我们用Ar~+激光514.5nm照射的结果相似。本文还就激光对造血细胞的作用机理作了初步探讨。
