Objective To investigate the feasibility of actA gene as a marker for lineage classification of Listeria monocytogenes(Lm) and the genetic and evolutionary relation between Chinese strains and US strains. Methods A total of 40 Lm isolates were obtained from food (n=32) and sewage (n=6) samples as well as from two human listeriosis cases that occurred in China. All isolates were characterized for molecular subtype by DNA sequencing of the 597?bp 3′ terminal region of the virulence gene actA. Sequence data were used to classify the 40 Chinese Lm isolates into sequence types and phylogenetic lineages and to compare the sequence types of the Chinese isolates with those of isolates from the US. Results The Chinese isolates could be separated into two genetic lineages with 14 and 26 isolates belonging to lineages Ⅰ and Ⅱ, respectively. Lineage Ⅱ could further be subdivided into two clusters (ⅡA and ⅡB). Lineages Ⅰ and Ⅱ were identical to the two lineages previously described among US Lm isolates. A total of 14 actA sequence types could be differentiated among the 40 Chinese Lm isolates; two specific actA sequence types were found from both Chinese and US isolates. Conclusion DNA sequencing of 3′ terminal region of actA is an effective method for the fast subtyping and lineage classification of Lm. Lm clonal groups found in China are very similar to those found in US. Many Lm strains may thus represent globally distributed clonal types.

目的 探讨产单核细胞李斯特菌(Lm)的actA基因3′末端核苷酸序列作为其遗传谱系分析的可能性,及中国与美国的Lm分离株遗传进化的关系。方法 采用特异性引物扩增40株分离自食品、污水及临床患者的菌株actA基因3′末端597 bp的核苷酸序列,所获得的DNA序列用于Lm遗传谱系的建立并将其分为不同的actA基因型;比较中国Lm 分离株与美国分离株actA基因型与遗传谱系的差异。结果 40株Lm中国分离株可分为两个遗传谱系(谱系Ⅰ和谱系Ⅱ),其中谱系Ⅰ包括14株,谱系Ⅱ包括26株。26株谱系Ⅱ的菌株又可被分为两个亚群(ⅡA和ⅡB)。与15株已发表的美国分离株序列比较发现中国分离株与美国分离株的谱系相一致,在40株中国分离株中有14个actA基因型,其中有2个与美国分离株的actA基因型相同。结论 actA基因3′末端核苷酸序列适合于Lm的谱系分析,且中国分离株与美国分离株具有较大的相似性,Lm 的克隆组群可能在全球范围内存在。

The RAPD markers are used to analysis the population genetic structure of rotifer Brachionus quadridentatus in Shuiguo Lake and Guozheng Lake.The results show that the genetic diversity within clonal groups is much higher than the genetic diversity among groups and the genetic structure between the two lakes is consisted of randomly distributed individual with large genetic difference.
