Immobilized enzyme can be reutilized to synthesize α ZAPM through batch reactions.

以固定化嗜热菌蛋白酶催化合成天苯二肽前体α- ZAPM,再脱去保护基后得到天苯二肽。固定化优化条件为 :海藻酸钠起始浓度 4 % ,戊二醛 0 .5% ,交联时间 10 min,凝胶球直径 2 .0至 2 .5mm。固定化酶最适反应 p H为 7.0 ,最适反应温度为 30℃。固定化酶对 p H和温度的稳定性明显提高 ,可连续进行批式反应

Papain-catalyzed synthesis of aspartame precursor Z-APM in biphasic solvent system wasinvestigated. Z-APM was obtained in the yield of 62.8% under mole ratio of substrates 1:1, temperature37℃ and reaction time 72h. The result shows that the catalytic activity of papain in n-BuOH-H2O wasbetter than in other organic solvent-H2O systems.

本文报道在两相溶剂体系中利用木瓜蛋白酶催化合成Aspartame前体Z-APM的研究。实验结果表明,木瓜蛋白酶在水-正丁醇两相体系具有较好的催化活性,在底物比(Z-L-Asp:L-PheOMe)为1:1,温度37℃条件下连续反应72h,Z-APM的产率可达62.8%。
