The research of viroid in plant tissue culture was just begun in the world. Cultivation of viroid in tomato cells has been reported in recent years. Gynura aurantiaca was inoculated with the leaf sap or crude extract containing nucleic acid from leaves of Cuba Mosaic Orange or Bing-Tang Orange infected by citrus exocortis viroid ( CEV ) . About 1 cm2 sized pieces of young leaves or auxilliary buds used as explants were sterilized and then cultivated on slightly modified B5 or MS semi-solid medium at 24-27℃...

The research of viroid in plant tissue culture was just begun in the world. Cultivation of viroid in tomato cells has been reported in recent years. Gynura aurantiaca was inoculated with the leaf sap or crude extract containing nucleic acid from leaves of Cuba Mosaic Orange or Bing-Tang Orange infected by citrus exocortis viroid ( CEV ) . About 1 cm2 sized pieces of young leaves or auxilliary buds used as explants were sterilized and then cultivated on slightly modified B5 or MS semi-solid medium at 24-27℃ under shade condition. After induction for 2 to 3 weeks in the medium, calli appeared, some loose and fragile like "white edible fungis", others grey black and rigid like "tumour". The results of bidirectional PAGE followed by silver staining and dotblot hybridization specially applied to detect viroid showed that not only the calli could form from explants of CEV-infected young leaves and buds but also CEV may be able to amplify in them. These results suggested that tissue culture system of Gynura aurantiaca for the cultivation and study of CEV has preliminarily been establishehed in our labaratory.

用含裂皮病类病毒(Citrus exoeortis viroid,简称CEV)的古巴花叶橙、冰糖橙的叶汁和核酸粗提物感染爪哇三七(Gynura aurantiaca),取其腋芽和嫩叶作外植体,经消毒处理,接种在稍加改良的B5和MS半固体培养基上,置于24℃~27℃遮光培养,经2~3周诱导形成愈伤组织,质地有的松散易碎似白木耳状,有的坚硬灰黑似多瘤状,经双向凝胶电泳-银染法和点杂交法检测,证实用CEV感染的嫩叶和腋芽作外植体,不仅能诱导形成愈伤组织,而且CEV还可能在愈伤组织中复制,结果表明,已初步建立了爪哇三七的组织培养和研究类病毒的离体培养系统。

利用根瘤土壤杆菌Ti质粒转化系统,采用叶盘法将CMV-Cp基因转入番茄细胞中,获得42株转基因植株,经Southernblot和Westernblot检测,证明CMV-cp基因已进入番茄核染色体中,并能正常表达。通过对转基因植株R1和R2代苗期人工接种CMV鉴定,发现CMV-Cp基因产物对CMV有一定抗性,其抗性遗传符合孟德尔一对基因控制性状的遗传规律并能稳定遗传。

There was always a difference inoxidative burst between the two nearisogenic tomato cell cultures and the relative light units in resistant cell cultureexhibited by luminol chemiluminscencewas 100 to 1000 times that in the susceptible one (Figs. 1, 2 ). The dependence of oxidative burst on concentration can be expressed by a sigmoidcurve(Fig. 4 ). When the concentrationof the elicitor was less than 1 μg/ml,the level of oxidative burst was verylow. It increased rapidly with the concentration the elicitor and up to a maximium value of relative light units between 25 to 30 μg of elicitor per ml ofcell suspension at 25 min.

从对Verticilliumalbo-atrum感抗性不同的两株近等位基因番茄获得细胞悬浮培养物S-GCR26和R-GCR218。用.albo-atrum激发子处理后,敏感性的S-GCR26未表现出明显的反应性氧改变。抗性的R-GCR218则响应以反应性氧迸发。反应性氧迸发中的细胞,其反应性氧累积量与激发子浓度的关系呈近S形。悬浮介质、某些无机离子、培养物的生长状况和培养日龄等因素均影响细胞对激发子的反应性氧迸发响应。
