Purpose To study the significance of quantitative pathological analysis on bone marrow tissue of hematological diseases. Methods Fifty nine bone marrow tissue sections from different hematological patients and six from normal persons were analyzed on hemopoietic tissue, adipose tissue, bone trabecula by quantitative methods, massons trichrome stain and Gomoris silver stain. Purpose To study the significance of quantitative pathological analysis on bone marrow tissue of hematological diseases. Methods Fifty nine bone marrow tissue sections from different hematological patients and six from normal persons were analyzed on hemopoietic tissue, adipose tissue, bone trabecula by quantitative methods, massons trichrome stain and Gomoris silver stain. Results In aplastic anemia group the number and area of hemopoietic tissue and bone trabecula were signficantly decreased ( P<0 05,P<0 01 respectively), while the number and area of adipose tissue were significantly increased ( P <0 05) compared with bone marrow tissues from normal and other diseases. Massons collagenous fibers were positive in hematological disease groups, and its moderate and strong positive fibers were higher in acute leukemia (28%) and MDS (67%) groups than those in chronic aplastic anemia group (13%). In MDS group, 78% cases with moderate and strong positive Gomoris stain were found, while there was no positive staining in acute leukemia and aplastic anemia groups. Conclusions Quantitative pathological analysis on bone marrow tissue has a differential diagnostic significance in hematological diseases.

目的:研究骨髓组织病理学定量分析在血液病中的作用。方法:对59例血液病患者及6例正常人骨髓组织切片进行了造血组织、脂肪组织及骨小梁等病理学定量分析;Masson三色染色和Gomori网状纤维染色观察。结果:与其他血液病及正常人骨髓相比,再生障碍性贫血(再障)病人骨髓的造血组织数及造血面积明显减少(P<0.05),脂肪组织数及脂肪组织面积明显增多(P<0.05),骨小梁数及骨小梁面积明显减少(P<0.01)。Mason胶原纤维在血液病中均有不同程度的阳性,其中急白组和MDS组高于其他组。AL组中度阳性及强阳性为28%,而MDS组为67%,CAA组为13%;Go-mori网银蛋白染色在急白组无中度阳性和强阳性,在MDS组中为78%,而再障组无中度阳性和强阳性。结论:骨髓组织病理学定量分析对血液病具有鉴别诊断意义。

Objective:To isolate and establish long term T cell clones derived from bone marrow(BM) of a patient with severe aplastic anemia(sAA).Methods:CD34 + cells and CD3 +T cells were enriched from BM aspirates,respectively by positive and negative immunomagnetic sorting protocol.BM of exicised rib of a non hematological patient was used as control. Irradiated CD34 + cells together with T cells were subject to one way autologous mixed lymphocyte culture(AMLC).Viable T cells surviving from AMLC were limit diluted and single cell was plated into microtitier wells along with feeder cells.Then all wells were screened for the growing cell wells and they were transferred into new wells to be expanded and maintained.Each clones obtained were primarily assessed CD4/CD8 phenotype and their homogeneity by immunocytochemical analysis.Results:The purity of sorted CD34 + and T cells was more than 90%. On AMLC,a total of 4×10 3 cells were viable with sAA,but 65 cells viable with control.Upon single cell seeding it 11 growing cell wells were found for sAA,and no one growing cells for control.The actual plating rate(2.1%) was within Poisson theoretic value range (<26%),which means a successful cloning procedure A primary analysis of these 11 clones showed 9 clones expressed CD4 homogeneously,but the other 2 expressed CD8 homogeneously.Conclusion:11 T cell clones were established which seems reactive to CD34 +stem/progenitor cells of a sAA patient.Further studies on these clones should facilitate elucidating immune mechanism of sAA.

Objective:To observe white cell count of 21hematologic patient samples with WBC Optical Count (WOC) and WBC Impedance Count (WIC) with full-automatic blood cell analysis instrument Abbott CD3700. Method: Through measuring 100 normal human samples and 21hematologic patient samples, we acquired white cell count with WBC Optical count (WOC) and WBC Impedance Count (WIC), 15 and 9 times ordering number white cell count, and measurement rate of WBC. The data is divided into control group, patient group and lyse-resistant red cell (RRBC) group and then done statistic analysis. Result: There are statistic difference between WOC and WIC both control group and hematological patient group. There is obvious difference between two methods in the hematological patient group. The correlation between WBC Optical count rate and WBC impedance count is weak. However, there is no difference of WIC between patient group and RRBC group. Among of them, with extension of time preparing to handle, WBC Optical Count rate and measurement rate are lower. Conclusion: WBC Optical Count combined with WBC Impedance Count can effectively increase the accuracy of white cell count, and it also hints that suspicious sample is handled with handwork method.

目的 :使用雅培CD370 0全自动血细胞分析仪对 2 1份血液病患者样本检测中激光散射法 (woc)和电阻法(wic)白细胞计数进行观察。方法 :通过测定 10 0例正常人样本和 2 1例血液病患者样本 ,获得激光散射法和电阻法白细胞计数值及 15个和 9个时间点的白细胞数、白细胞检测率 ,将所获数据分为正常对照、患者和阻抗红细胞组进行统计学分析。结果 :正常对照和血液病样本woc和wic存在统计学差异 ,血液病样本两种方法测得值差异大 ;白血病样本中激光散射法检测率与电阻法检测率的相关性差 ,而患者组和阻抗红细胞组wic无统计学差异 ;其中 19例样本随预处理时间延长 ,激光散射法白细胞计数及检测率降低。结论 :激光散射法结合电阻法可有效提高白细胞计数的准确性 ,并提示对可疑样本进行手工方法处理
