It has become increasingly clear in recent years that various chemicals,clastogens andcarcinogens get into the human body and cause damage to DNA.In order to determinetheir effects,the SCE analysis has come to be a sensitive means of monitoring DNA damage.The SCE technique is a sensitive,simple and rapid method compared with the convention-al chromosome aberration analysis.Applying the SCE analysis to the non-pregnant and parturient pregnant women andtheir own neonates is significant for maternal and child health,perinatalogy,family planningand eugenics.The SCE analysis of normal non-pregnant women,parturient pregnantwomen and their neonates may be regarded as a basic research work in this field.The various samples of blood were obtained from 30 normal healthy nonpregnant womendonors and 30 normal parturient pregnant women.The samples of whole blood of the30 neonates were obtained from the umbilical cords.Add 0.25—0.3 ml of the whole blood to the culture(RPMI 16404 ml+fetal bovineserum 1 ml+proper amount of PHA and heparin)and incubate it for 24 hours at 37℃±0.5℃,then add BUdr,given a final concentration of 12 ug/ml,and again add colchi-cine with a final concentration of 0.04 ug/ml,continuing the incubation in the dark forthe last 4 hours.The average value of SCE of the normal non-pregnant women was 6.53 and that ofthe parturient pregnant women was 6.65,there being no significant difference between them(P>0.05).The SCE average value of neonates was 5.04,and it was significantly differentbetween the parturient pregnant women and their own neonates(P<0.01).

This study provided evidence that oxidation hair dyes or their metabolites may reach the germ cells of adult mice in sufficient amounts and the duration of their injurious actions was quite long to produce DNA damage. However, "Meirong" hair dye produced less damage to DNA than MMS, and did not affect development of the germ cells in male mice.

本文应用雄性小鼠生殖细胞非程序DNA合成(UDS)检测方法研究了“美容牌”染发剂对雄性生殖细胞DNA的损伤作用。发现该染发剂诱发的UDS与药物剂量呈线性正相关。提示此类染发剂对雄性生殖细胞DNA有损伤作用。其遗传毒理效应应引起注意。

The results showed that nitrofurantoin may induce an increase in abnormal sperms, and the early spermatocytes are the cell stages most sensitive to the production of sperm abnormalities by nitrofurantoin. The mechanism of induction may be a direct damage to DNA or an interference with the expression of this genetic material.

本文应用小鼠精子畸形试验方法,研究了呋喃妥因对雄性生殖细胞的损伤作用。结果表明该药对小鼠精子有明显的致畸作用,以精母细胞期对药物的作用最敏感。文中对精子畸形与雄性生殖细胞发育阶段的关系进行了简要阐述。
