Objective To study the isolation,characteristics and comparison of several detection methods of Helicobacter pylori.Methods Anaerobic cultivation and PCR were used to isolate and determine H.Pylori preliminarily.One strain was further determined by various tests of characteristics.Results About forty strains of H.pylori were isolated and preliminarily identified from gastric mucosa biopsy specimens in 106 patients presenting for gastroscopy at hsopitals.HP-WGM was gram-negative,curved bacilli which ha smooth coats,several sheathed single polar flagella.After 2~3 days'incubation,it grew evidently as about 1mm in diameter,round,protruding,uniform in edge,semi-translucent coloies on urea agar rolled-tubes incubated at 37℃ under microaerophilic condition.It was positive in urease,catalase,oxydase tests and negative in H 2S producing and gelatin liquefaction tests.It couldn't reduce NO - 3,could tolerate 3 5% NaCl and 1% glycine and hydrolyze starch.The organism is immunogenic.Conclusion HP-WGM could be used as an antigen to carry out ELISA for determining the extent of H.pylori infection.At least over two methods are necessary to diagnose the infection.But in scientific research,the culture and determination of bacteria is the most reliable way.

目的 分离一株幽门螺杆菌 ,研究它的生物学特性及免疫原性 ,同时比较几种检测手段的优势。方法 采用厌氧微生物培养技术及PCR检测方法分离、初步鉴定幽门螺杆菌。重点对其中一株菌HP -WGM做各项生理生化实验 ,ELISA试验分析。结果 从 10 6位患者中分离鉴定得到 4 0株疑似幽门螺杆菌。HP -WGM菌株革兰氏染色阴性、杆状、稍弯曲、菌体的一端着生数根带鞘鞭毛。该菌脲酶、触酶、氧化酶均为阳性 ,不还原硝酸盐 ,H2 S的生成试验阴性 ,其生长耐受 3 5 %NaCl、l%甘氨酸 ,水解淀粉 ,不液化明胶。该菌有一定的免疫原性。结论 HP -WGM菌株可作为一种抗原用于检测幽门螺杆菌的感染程度。对于疾病的确诊 ,应采用至少 2种以上的检测手段 ;用于科学研究时 ,细菌培养、鉴定的方法才是最为可靠的手段。

可导致败血症的细菌种类很多。针对不同细菌采用不同的抗生素进行治疗 ,是败血症能否治愈的关键。目前临床采用的细菌鉴定方法主要是应用全自动血培养仪及全自动细菌鉴定仪。实验室鉴定方法包括聚合酶链式反应、限制酶片段长度多态性分析以及生物芯片技术等。上述各方法均各有其优缺点。该文综述其优缺点并预测败血症细菌鉴定的未来研究方向
