Aim:To study the erythrocyte rheology during myocardial ischemia and reperfusion.Method:Free radicals,lipid peroxides(MDA),activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),red cell deformability,molecular fluidity of lipid and protein of red cell membrane were measured in 15 patients with atrial or ventricular septal defect.Blood samples were collected from coronary sinus during open heart surgery under cardiopulmonary bypass(CPB).Result:After myocardial reperfusion,the level of free radicals,MDA,activities of GSH-Px were significantly increased and activity of SOD was decreased.As consequence of peroxidation,the molecular mobility of red cell membrane were significantly decreased which induced increase in viscosity of red cell membrane.Conclusion:The changes of erythrocyte deformability may play a critical role in reperfusion injury during CPB.

对15例房或室间隔缺损病人在行体外循环心内直视修补术中,心肌缺血/再灌注期间的冠状静脉窦血中自由基水平、脂质过氧化物、自由基清除酶活性、红细胞变形性和红细胞膜脂质分子及蛋白分子流动性等指标进行了动态观察。结果发现,再灌注后的自由基和脂质过氧化物水平明显增高、血浆SOD活性降低及红细胞谷胱苷肽酶活性提高,同时,红细胞膜分子尤其是脂质分子流动性下降,从而导致整个红细胞变形性恶化,这是造成再灌注后微循环障碍的危险因素。

n order to investigate the effects of free radicals on RBCs rheology and the molecular fluidity of RBCs membrane, after reactions with free radical system, membrane elastic modulus, surface index of cells, rotational correlation times of lipids and proteins of RBCs membrane were studied in vitro. Results showed that all of the membrane shear elastic modulus, rotational correlation times of the membrane lipids and proteins were significantly increased by 49.8%, 49.5% and 191.4% separately after free radical attacking, but no influence on cell volume was found due to unchanged cells surface index. In the present study, the changes of rheological parameters were concentration depend on free radicals, and partly recovered three hours after damage. All the above demonstrated that the immobilities of both lipids and proteins resulted from free radical damage contributed together to disfluidity of RBCs membrane.

为探明自由基引起红细胞流变性改变的作用机理,我们对体外自由基作用三十分钟前后红细胞膜剪切弹性模量、细胞表面指数、膜脂质分子和蛋白分子流动性进行了比较分析。发现自由基作用后,红细胞膜剪切弹性模量、膜脂质分子和蛋白分子的相对旋转时间分别增加了49.8%、49.5%和191.4%,而红细胞表面指数无变化;此外,随着自由基浓度的降低以上变化也随之减轻;在自由基损伤后的第三小时,上述改变部分回复。本研究说明,自由基引起脂质和蛋白分子流动性降低共同导致红细胞膜剪切弹性模量的增加,并且一定浓度自由基对红细胞流变性的损伤是部分可逆的。

Objective:To investigate the molecular mechanism of Fructus Ligustri Lucidi (RLL) in protecting membrane rheo- logical characteristics of red blood cell from free radical damage.Methods:The changes in membrane molecular chemical structure and their fluidity after free radical attacking for 30 minutes were observed and compared to that of protected group by RLL.Results:In the RLL group:(1) The productive rate of free radical lowered for at least 64% in average; (2) The membrane elastic shear modulus increased for 27.7% only;(3) Protective effect on either membrane protein or lipid molecular fluidity was not significant;(4) RLL could prevent damage of hydrogen bond from free radical attacking but couldn't prevent membrane protein reconstitution from disulphic bond formation.(5) Peroxidation in either mem- brane hydrophobic or aqueous areas was not limited.Conclusion:Degree of membrane damage was related to the concen- tration of free radical,it could be partly restored after 3 hrs attacking,the recovery was dependent on free radical concen- tration.RLL could relieve the damage on cell membrane to a certain degree.

目的:为探明女贞子对自由基损伤红细胞膜流变性的保护作用的分子机理。方法:对自由基作用前后红细胞膜分子流动性和分子结构的改变与自由基加女贞子提取液共同作用后的红细胞膜相应参数进行了比较。结果:加入女贞子提取液后:(1)自由基产生率平均降低了64%以上。(2)红细胞膜剪切弹性膜量只增加27.7%。(3)对自由基引起的膜蛋白分子和膜脂分子流动性降低的保护不甚明显。(4)能够防止自由基对氢键的破坏,但不能防止二硫键形成所造成的膜蛋白结构的重构。(5)不能减轻自由基对膜磷脂亲水头部和疏水尾部的过氧化作用。结论:本研究中自由基损伤程度与自由基浓度有一定的量效关系,并在膜脂分子损伤后3h 表现出部分可逆性,但与自由基浓度相关。女贞子可减轻自由基对膜分子的损伤程度。
