Objective To observe the effect in repairing segmental radial bone defect of rabbits with different bone. Methods The 15 mm segmental bone defects were created on bilateral radius in 48 New Zealand rabbits and were divided into four groups. Group A received deep-low-temperature-frozen allogeneic bone combined with autologous bone marrow, group B: deep-low-temperature-frozen allogeneic bone, group C: autogenous bone, group D: fresh allogeneic bone. General observation, HE histological section, transmission electron microscope and X-ray were done in different stages. Results There were all bone formation phenomenon in four groups, Bone formation and synostosis of group A and C surpass group B , group B surpass group D obviously; Cell accrementition, cell nucleus dissociation , plentiful cytoplasm, nuclear envelope and abundant cell organ of group A and C surpass group B and D obviously; The bone defects healed in the 8th week in group A and C, in the 12th week in group B, but did not healed in the 12th week in group D. Conclusion The deep-low-tem-pera-ture frozen allogeneic bone combined with autologous bone marrow (ABM) of rabbits has favourable his-tocompatibility and osteogenesis; It's possible to repair bone defects with deep-low-temperature-frozen allogeneic bone combined with ABM displacing autogenous bone.

目的对比观察不同骨移植材料对兔桡骨节段性骨缺损的修复效果。方法48只新西兰大白兔采用桡骨15mm节段性骨缺损模型,随机分为4组,每组12只,A组植入深冻异体骨复合自体骨髓,B组植入深冻异体骨,C组植入自体骨,D组植入新鲜同种异体骨。术后不同阶段分别行组织学、透射电镜及X线检测。结果四组骨缺损均有成骨现象发生,骨生成、骨连接情况A、C组优于B组,B组优于D组;A、C组细胞增生活跃、核呈分裂相、胞质丰富、核膜光整、细胞器丰富,同比均优于B、D组;骨缺损愈合时间A、C组为8-10周,B组为12周,D组骨缺损在术后12周仍未愈合。结论提示兔深冻异体骨复合自体骨髓具有良好的组织相容性及成骨作用,其取代自体骨植骨修复骨缺损具有可能性。

OBJECTIVE: To probe into the development of microinvasive bone restorative procedure according to recent researching status of injected bone restorative materials in the treatment of bone disunion. DATA SOURCES: A computer based search of Medline for literatures related to bone nonunion, bone induction, injected materials and bone restoration from January 1985 to December 2005 was conducted with the of "fracture nonunion, bone induction,carrier, percutancous", and limited the language to English. Meanwhile, CBM was searched for articles related to injected bone restorative materials from October 1994 to October 2004 with computer with the key words of "fracture nonunion, bone induction, carrier" and limited the language to Chinese. STUDY SELECTION: Data were searched firstly, literatures about bone-induced materials and bone nonunion were selected, full-text was looked for. Accepting criteria: ① Bone nonunion and bone induction. ② Injected materials and method of bone formation. Excluded criteria: ① Infection of bone, bone benign tumor and bone malignant tumor. ② Reviews, repetitive studies and Meta analysis articles. DATA SYNTHESIS: Injected bone restorative materials were widely applied in the treatment of clinical late healed and disunion of bone fracture, which was characteristic by simple, economic, effective and with less complications. Of the total, autologous bone marrow (ABM) and compounds could promote the bone healing though the differentiation of matrix cells and the sythesis of bone matrix. As an effective induced factor of bone formation, bone morphogenetic protein (BMP) and complex had effects on mesenchymal or marrow stroma cells (MSC), tured them into cartilage or bone cells, which had good characteristic of bone induction. Calcium acid phosphate cement and complex, as an bone-conduction materials, were mainly considered as the slow-release carrier of bone induction to form the compound bioactivity materials, which was endowed with ability of both bone conduction and bone induction were significantly enhanced the restorative effect on bone deficiency. Bone matrix, bone matrigel, collogen (mainly type I collogen) and complex had double function of conducting and inducing bone formation, of which if combined with other better mechanics-nature materials, better effects would be achieved. Research in the laboratory has proved the fibroblastic potency of bone formation, while the combined applying of many bone growth factors would has much better effects in promoting the bone formation. But there were some problems still existed in the clinical application of exogenous growth factors such as factor sources, cost, immune reaction, side effects and so on. Botryomycosis as a proliferating agent of micrangium, could sitimulate the micrangium grow towards the ends of bone fracture, and widely applying in the treatment of delay healed and nonunion of bone fracture in recent years. However, satisfing effects of parenteral solution of cord blood and placenta on aspect of promoting the bone formation had been achieved. CONCLUSION: Injected bone restorative materials has a wide prospect of clinical application, while the indicatios and fixed ways should be in strict control. Problems such as effectiveness and persistence of injected materials, selection of carriers, ectopic ossification of transplanted sites and so on needs further researching and settlement.
