To evaluate the efficiency and safty of cottonseed (gossypol<0.05%)contain- ing Bt-protein from Bt-transgenic cotton plant resistant to insect,as feed for fish,a 12- week chronic toxicity test was conducted with Cypinus carpio using diets containing 30%(by weight) processed cottonseed meal from the insect resistant and normal cotton plant. No deleterious effects on carp growth and development were observed by investigating be- havior,weight gain rate,feed efficency and survive rate of fish,No significant differences(P<0.05)in the blood indexes such as WBC、Hb,GPT,BUN and the rate of micronucle- us of red corpusles were found.NO pathological changes were found in organs such as liv- er,kidey,intestines,heart ect.of carp in both test groups and control groups byhistopathology examination,Also,no differences(P>0.05)were observed in the organrelative weight between test groups and the control groups. These results demonstrate thesafety of the cottonseed from Bt-transgenic cotton plant to water animals when used asfishery feed resources.

为评价转Bt-基因抗虫棉脱酚后棉仁粉用作鱼饲料的有效性和安全性,本文进行了12用的鲤鱼慢性毒性试验。对鲤鱼的增重率、特定生长率、饵料系数、成活率等营养学指标的观察以及对鲤鱼的血像、生化、遗传、病理组织等毒理学指标的分析结果表明,就上述所观察的指标,含抗虫棉棉仁粉和含泗棉3号棉棉仁粉之间均无显著性差异。证明转Bt-基因抗虫棉棉仁粉经脱棉酚处理后,对水生动物无毒性作用,与常规棉棉仁粉一样可用作水生动物饲料的蛋白质资源。

In a 28 day feeding test with rats and a 8 day feeding test with quail,no adverse effects on rats or quails were observed in terms of animal behavior,weight gain and feed utilization rate.In addition,no pathological changes were found in organs such as liver,kidney,intestines and testes of rats in both test and control groups by histological section examination.No signifcant differences (P>0 05) were observed in organ relative weight, GPT activity and urea nitrogen in the blood of rats between the test groups and the controls.These results demonstrate the safety of low gossypol cotton seed from Bt transgenic plants used as feed resources.

经小鼠急性中毒试验确定,转Bt-基因抗虫棉棉仁粉属无毒级。同时,进行了28天大鼠喂养试验和8天鹌鹑喂养试验。结果显示:各试验组体重、食物利用率与对照组无显著差异,受试动物生长发育及行为正常,无死亡。对大鼠肝、肾、胃、盲肠、结肠、小肠及睾丸进行组织切片检查,均未见病理性改变。大鼠肝、肾、睾丸的重量比及血液中谷丙转氨酶活性和尿素氮水平均在正常值范围内,试验组与对照组无明显差异。上述结果表明,转Bt-基因抗虫棉棉仁粉经脱棉酚处理后,对动物无毒害作用,因而与常规棉棉仁粉一样,可作为畜禽饲料资源

By injecting a synthetic Bt gene (GFM CryIA) cloned in a plant expression vector, pBI121 1, into ovaries of cotton varieties “Zhongmiansuo 19”,“Huanong 916”,“541” and “3517”, 29 Bt transgenic plants were obtained,which had high bollworm larvae mortalities (over 80%), and showed PCR positive in proof of the integration of the Bt gene into the cotton genomes. The fact that as many plants as 26 in 29 of the Bt positive ones had killed bollworm larvae at the death rate of 100% indicated that the fully modified Bt gene succeeded in raising Bt proteins toxicity to bollworm.

采用子房注射法,将构建在植物高效表达载体上的全序人工合成的Bt杀虫基因GFMCryIA转入棉花品种(系)“中棉所19”、“华农916”、“541”和“3517”中,获得对棉铃虫幼虫致死率在80%以上的植株29株。用PCR法检测,结果表明这些植株均呈阳性。在这29株阳性植株中,对棉铃虫致死率为100%,且自身只有轻微伤害的植株有26株。这说明Bt基因经改造,增加其提高转录与翻译能力的表达调控元件后,对棉铃虫的毒杀作用明显提高。
