A method for the determination of xylose and xylitol in fermentation broth by high performance liquid chromatography is described. 建立高效液相色谱检测发酵液中木糖和木糖醇含量的分析方法。色谱柱为HypersilNH2 柱 (4 .6mmi.d.× 2 5 0mm ,5 μm) ,柱温 3 5℃ ,流动相为乙腈—水 (80∶2 0 ) ,流速 1 .0mL .min 1,示差折光检测器检测。木糖和木糖醇在 3 .0~ 60mg.mL 1范围内 ,峰面积与其浓度线性关系良好 (г=0 .9995 ) ;平均回收率分别为 96.0 7% (n =5 ,RSD =0 .5 1 % )和 97.47% (n =5 ,RSD =1 .1 3 % )。方法简便、快速、准确。

The paper’s aim was to get the optimum wavelength for the determination of xylose content by DNS method. The reaction solution of xylose and DNS regent was scanned from 400~700nm,and the OD values were determined, respectively. The results showed that the max absorption is from 480~490nm, and the line range of the method is 150~600μg/ml (r=0.99997) xylose at 480nm. The result also showed the sensitivity, accuracy, precision are the highest at 480nm, and the recollection rate is more than 99.5%....

为了正确地选择DNS法测定木糖含量时的最佳测定波长,扫描了木糖与DNS试剂反应液的紫外-可见吸收光谱(400~700nm),并分析测定了其在480、500、520、540、550、560nm时的OD值。结果表明:木糖与DNS试剂反应液的最大吸收波长在480~490nm;在480nm波长下测定时,灵敏度最高,且于150~600μg/ml范围内呈现较好的线性关系,其回归系数r值=0.99997,回收率大于99.6%。在500、520、540、550、560nm测定时,灵敏度依次降低。因此,DNS法测定木糖时的最佳测定波长为480nm。
