采用聚合酶链反应(PCR)和限制性片段长度多态性分析(RFLP)技术,对4l例大肠癌组织大肠癌缺失基因杂合性缺失进行研究。结果表明,38例大肠癌信息个体中有21例(55.3%)发现DCC基因杂合缺失。DCC基因杂合性缺失与肿瘤大小、组织学类型和浸润深度无关,但有淋巴结转移(80.00%)和DukesC、D期(71.4%)大肠癌Dcc基因杂合缺失率显著高于无淋巴结转移(39.1%)和DukesA、B期组(35.3%,P<0.05)。结果提示,DCC基因杂合缺失多发生于大肠癌的晚期,DCC基因杂合性缺失的检测有可能成为大肠癌病人预后评价的指标。

These results suggest that LOH at the DCC locus occurs in late stage of the disease and detection of LOH at the DCC locus may be useful for predicting the prognosis of the gastric cancer patients.

为了解DCC基因改变在胃癌发生发展中的作用及与临床预后的关系,本文对45例手术切除胃癌组织DCC基因杂合性缺失进行分析。结果45例胃癌中发现杂合缺失15例,缺失率为33.3%。杂合缺失与临床分期相关,但与组织学类型、肿瘤大小、浸润深度、淋巴转移无明确关系。以上结果提示,DCC基因缺失是胃癌的晚期改变,检测其杂合缺失对判断病人的预后可能有一定帮助。
