Determination of lipid signaling pathways and the production of some intracellular signaling molecules has been an focus of some research work in the field of signal transduction research. For example, the determination of the regulation of different phospholipase activity and the formation of intracellular second messengers and other lipid bioactive molecules from different sources is an important component in the studies on the biological effects and their mechanisms of growth factors and other molecules. Here, a method was introduced for selectively labeling different phospholipids, which has been widely used in the study of lipid signaling pathways. Experimental evidence was presented to demonstrate how to determine the different signaling pathway and the generation as well as the alteration of relevant signaling molecules. The application of this method is flexible and has a high reproducibility. It can be used to pinpoint some signaling pathways and the formation of different signaling molecules. A brief introduction about lipid signaling and its significance in the area of signal transduction was also given.

Results\ The fusiform fibroblasts oriented radiantly or longitudinally and closely packed onto the HA ECM, they attached firmly and proliferated to confluence on the stromal surface of HA ECM. Conclusion\ The optimal cell concentration is 3 5×10 6/ml, HA ECM is ideal carrier for fibroblasts because of its excellent scaffold and diffusion character istics. Futhermore it has the bioactive molecules such as fibronectin and laminin which play an important role in fibroblasts attachment and proliferation on HA ECM\;

目的 通过人羊膜细胞外基质 (HA ECM)与成纤维细胞体外培养 ,观察成纤维细胞在HA ECM上的生长特性 ,以及胶原原纤维产生情况 ,为使HA ECM成为皮肤组织工程化的载体打下基础。方法 将不同浓度的成纤维细胞种植于HA ECM上进行体外培养 ,通过倒置显微镜、光镜及透射电镜 ,观察细胞在HA ECM的生长、排列 ,以及细胞的粘附和胶原原纤维产生情况。结果 梭形的成纤维细胞在HA ECM上生长良好 ,呈放射状或平行排列 ;组织切片发现HA ECM上有较多梭形细胞附着 ,细胞排列紧密 ;电镜下见成纤维细胞胞膜基底部向HA ECM内伸出数个突起 ,细胞与羊膜基质粘附相当牢固 ,胞外有较多胶原原纤维堆积。结论 种植于HA ECM的成纤维细胞有一个最适宜的细胞浓度 (3 5× 10 6 个 /ml) ;HA ECM具有良好的支架和渗透作用 ,成纤维细胞在其上生长、粘附良好 ,胶原合成、分泌旺盛 ,HA ECM是一种较理想的成纤维细胞载体
