pEHT9 was constructed from pUR528, and used for the construction of plant thaumatin expression vector pBIT7. Then pBIT7 was introduced to Agrobacterium through direct transformation. Subsequently tobacco was transformed through Agrobacterium -mediated system. It has been confirmed that thaumatin gene has integrated into the genome of transgenic tobacco by PCR, PCR-Southern and Southern blot analyses.

Its ability to secrete these proteins is appealing for the production of heterologous proteins in the field of the genetic engineering.For the development of transformation system in A. oryzae, argB mutants were isolated by using UV irradiation. Transformation was achieved with the plasmids which carries the A. nidulans argB gene by using CaCl 2 mediated chemical method...

Its ability to secrete these proteins is appealing for the production of heterologous proteins in the field of the genetic engineering.For the development of transformation system in A. oryzae, argB mutants were isolated by using UV irradiation. Transformation was achieved with the plasmids which carries the A. nidulans argB gene by using CaCl 2 mediated chemical method and electroporation. The transformed plasmids were integrated into the host chromosome with multiple copies and stable transformants obtained. The frequency of transformation was, however, low between 10 to 50 transformants per μg of input DNA. Although the A. nidulans argB gene is not significantly homologous to the A. oryzae argB gene, it complements the A. oryzae argB mutant. In electric field mediated transformation, electroporation, the optimum conditions for A. oryzae was obtained at 6 3 kV/cm and 1540 ohm. The survivals of protoplasts in the electric fields were about 80% of nonprotoplasts at 1 3 kV/cm to 50% at 6 3 kV/cm. For the heterologous gene expression in A. oryzae, the plant originated nature sweeterner, thaumatin gene was introduced into A. oryzae argB mutant by using cotransformation with A. nidulans argB gene as nutritional selectable marker.An immunoreactive 22 kDa protein of a molecular weight similar to purified mature thaumatin was observed in the culture medium.It shows that the plant signal sequence directs thaumatin secretion in A. oryzae.
