1. The quail was capable of being experimentally infected by a virulent strain of fowl pox virus. A prolonged passage of this pox virus strain in quail has been successfully conducted and after 50 quail passages its infectivity for chickens has been sufficiently attenuated. 2. The infective titre of this quail-modified pox virus strain to chickens has been kept at a satisfactory level of about 10~(-4)-10~(-5) which is 2 logs lower than that of the initial virulent fowl strain. 3. The quail-modified pox virus was capable of growing in chicken embryos and causing mortality to infected egg-embryos after 8 successive egg passages, but neither its pathogenecity nor immunogenecity for the chickens has been altered. 4. The minimum immunizing dose (MID) of this viral vaccine prepared from the infected chick embryo CAM was 10~(-4)(by wing-web stick method), and the practical vaccination dosage of this vaccine corresponds to 100 MID. 5. The duration of immunity induced in fowl following a single vaccination lasted 4 months, and a partial immunity still existed after 6 months. 6. The keeping quality of the freeze-dried vaccine with definite virus titre have been demonstrated to be at 30℃ for 40 days, 20℃ for 90 days, 4℃ for a year, and-15℃ for 1.5 years. On the other hand, in the case of glycerinized vaccine, the keeping quality has suffered a considerable deterioration after 2 months' storage even at 4℃

按通过异种动物可以减弱病毒对原宿主的致病力而能较好地保持其优良抗原性的原理,选择了鹌鹑减弱鸡痘病毒。将自然鸡痘病毒102株感染成年鹌鹑的皮肤,连续传代适应后,可规律地产生局部皮痘反应,重者可散发全身痘,并可继续再适应于鸡胚,产生绒毛尿膜病变和痘斑。检验鹌鹑传代痘毒对易感鸡的致病力,证明在通过鹌鹑50代之后,其对鸡的致病力已明显减弱。系统检查不同代鹌鹑痘毒对鸡的安全性,通过强毒攻击试验,证明对各种日龄鸡的免疫性是坚强的。选用第162—200代以后的鹌鹑化鸡胚弱毒作种毒,制取甘油疫苗和冻干疫苗。本文报导这疫苗毒株静脉接种(刍鸟)鸡的安全性,疫苗的免疫试验、免疫期和保存性数据;还有在五个地区鸡场区域试验的显著免疫效果。并初步验证,适应机械化养鸡场应用气雾免疫也有良好免疫效果。现已经农业部制定制造检验规程,交生药厂投产推广应用。

To improve virus titers and stability of the lyophilized live viral vaccines and increase the production output of vaccine, some components, such as gelatin、sorbitol、saccharose、dextran and arginine,were screened for their suitability in using as stabilizers.The results showed that several stabilizer combinations can be used as future candidates upon further optimized.The selected stabilizer,compared with current production stabilizer,could give a vaccine output of 3 times higher.

The advent of cell fusion and recombinant DNA technlolgies has resulted in a burgeoning number of applications for animal cell culture technology. At present the industrial productions of the therapeutic biologicals(such as viral vaccines,recombinant proteins,and monoclonal antibodies)become the main driving force for the development of large-scale bioprocesses using animal cells. In addition to providing the way how to produce the required quantities of these biologicals effectively,the large scale animal cell culture technology and the manufacturing processes must also be designed to ensure that the products have appropriate quality for human therapeutic use and are free of potentially harmful contaminants,especially viruses and DNA. Through the considerable effort having been paid during the past decade, the manufacturing methods are now available to meet the above requirements.

细胞融合与重组DNA技术取得的巨大成功为动物细胞培养技术开辟了许多新的领域。目前各类具有明确疗效的生物制品(如病毒疫苗、基因工程蛋白质和单克隆抗体等)的工业化生产已成为动物细胞培养技术发展的主要动力。动物细胞大规模培养技术的研究开发,除了要解决如何高效地大量生产这些药物之外,其生产过程必须确保产品质量能适合人体治疗,且不含有害物质,尤其是病毒和DNA。经过十几年的努力,符合上述要求的生产方法已初步建立,本文拟就此作一简要综述。
