Molecular breeding techniques such as transgenesis, gene marker-assisted selection breeding and molecular marker-assisted selection breeding are potential approaches to developing new strains with enhanced disease-resistance in aquaculture, which should be strengthened in China. The aim of this paper is to provide a survey on the progress in the field of fish disease resistance-related functional genes and molecular breeding.

Random Amplified Polymorphic DNA (RAPD) technique was applied to investigate the WSSV-resistance molecular markers of Fenneropenaeus chinensis based on the samples after four-generation-selected breeding successively (XY(4)) and samples of the same origin without selected breeding (HB) in order to supply the theory and technology supports for the protection of Fenneropenaeus chinensis germplasm resource and molecular-marker-assistant breeding.220 single random primers and 114 pair of primers were used and a total of 2 439 reproducible RAPD markers were obtained in which 5 were special according to the band frequency and variety rule appeared in the samples.The 5 special genetic markers were sequenced and BLAST analysis showed no high similarity to the published function genes,and the same low similarities were revealed by the Amplified Fragment Length Polymorphisms (AFLP) technique based on the four selective breeding generation samples of Fenneropenaeus chinensis.Due to artificial selection,it was indicated that these special markers were tightly linked to WSSV-resistance trait but were not functional genes.

采用随机扩增多态性DNA(RAPD)技术对经过连续4代选育的抗白斑综合症病毒(WSSV)中国明对虾(Fennerope naeuschinensis)(XY4)及未经选育的与XY4来源相同的中国明对虾野生群体(HB)进行分析,以期筛选出与抗病性状相关的分子遗传标记,为进一步的基因定位以及中国对虾的种质资源保护和分子标记辅助育种提供有力的技术支撑和理论依据。共进行220个RAPD单引物和114对双引物的检测,产生标记数目共计2439个。依据标记在群体中出现的频率和变化规律,共筛选出5个可能与抗病相关的特异性标记,对这些特异性标记进行测序并将测序结果进行BLAST分析,发现测得片段的序列与数据库中序列的相似性较低,未能找到与所测序列同源性较高的功能基因。这与利用AFLP技术分析的结果一致。由于选育过程施加人工选择,推论这些特异性标记虽不是抗WSSV分子标记,但可能与抗病性状密切相关。
