It is important and valuable in application to construct variety fingerprint electrophoregram for the variety purity test, identification and management of variety based on variety's intrinsic and stable characters. Three primary fingerprint electrophoregrams of main maize hybrids and their parents inbred lines in Xinjiang were constructed based on EST isoenzyme, stored protein electrophoresis and RAPD in this paper. The results indicated that there were obvious differences in one of electrophoregrams among the materials used in the experiment. The three kind profiles constructed in the paper can apply the intrinsic and stable characters for the 8 maize hybrids and their parents inbred lines, and can also be served as an standards or basement for identification the variety or purity test more precision.

品种稳定、内在的特征,可供种子纯度检验及品种鉴定与管理,构建品种的指纹图谱具有重要的现实意义。采用脂酶同工酶、种子贮藏蛋白电泳及RAPD技术,初步建立了新疆玉米主要推广杂交种及其亲本自交系的三种指纹图谱。结果表明,种子脂酶同工酶、种子贮藏蛋白电泳谱带,在各供试品种与其亲本自交系间其图谱均具有较大差异,供试的24个材料间RAPD多态性差异明显。此三种图谱均可作为供试材料稳定、内在的特征,为准确地鉴定品种纯度及品种认定提供参考和标准,具有一定应用前景。

Isozyme and seed storage protein were insufficient number of polymorphic loci, which could not differentiate the varieties with close pedigree relationships. As isozymes are the products of gene expression, they may vary in different tissue and developmental stages. DNA - based markers can be effectively used in variety purity test by analyzing the differences of the nucleic sequences of varieties. As DNA - based markers are relatively unlimited and developmental stable, and the techniques can be economically, rapidly, efficiently and automatically employed , they have been being shown great potential utilization in variety purity test of maize.

品种纯度检验是玉米新品种保护及种子生产的技术依据,对玉米生产具有重要影响。本文综述了玉米品种检验的方法与技术,并对其进行了初步评价。研究认为形态鉴定法可之利用的形态性状较少,受环境条件影响较大,准确性较差。同工酶电泳和种子贮藏蛋白电泳技术由于同工酶位点和种子贮藏蛋白的类型有限,难以区分亲缘关系较近的品种。DNA分子标记技术通过鉴定DNA水平上的差异来检验玉米品种纯度,不受环境因素影响,可以区分表型上难以鉴别的品种。DNA分子标记技术简单、快速、易于自动化操作,在品种纯度检验上展示了广阔应用前景。
