Background & Objective: Esophageal squamous cell carcinoma is characterized by high incidence and high mortality. This study was designed to identify the proteins dysregulated in ECSS by. Methods: Microdissection of routinely unstained frozen sections was used to procure cancer cells from seven esophageal squamous cell carcinomas, and esophageal epithelial cells from normal tissues adjacent to the tumors. By two dimensional electrophoresis, protein profiles of normal and cancerous tissues were obtained. Selected proteins with altered expression in tumors were identified through MALDI TOF MS. Results: Compared the protein profiles of tumors with those of normal epithelia, 11 proteins were found as dysregulated in tumors. Among them, HSP27 was observed up expressed and three isoforms of ANNEXIN I were seen down expressed in cancerous tissues. Conclusion: Two dimensional electrophoresis coupled with mass spectrometry can contribute to rapidly screen dysregulated proteins in human maligancies. Altered expressions of HSP27 and ANNEXIN I were the frequent events in Chinese esophageal squamous cell carcinomas.

背景与目的:食管鳞状上皮癌(esophagealsquamouscellcarcinoma,ESCC)是一种具有高发性和高死亡率疾病,本研究应用蛋白质组学方法分析ESCC中异常表达的蛋白。方法:对ESCC/癌旁组织标本的冰冻切片进行显微切割,分别分离肿瘤细胞和癌旁上皮细胞,提取细胞总蛋白,通过双相电泳得到ESCC及癌旁上皮的蛋白表达谱,采用质谱技术鉴定肿瘤表达异常的蛋白。结果:比较7对食管癌/癌旁组织的蛋白表达谱,并对差异的蛋白进行质谱分析发现:11个蛋白在食管癌中表达明显异常,其中热休克蛋白27(HSP27)在3例ESCC中表达上调,钙依赖磷脂结合蛋白ANNEXINI在正常食管粘膜存在分子量基本相同、等电点彼此相差约为1的3种变体(isoforms),3种ANNEXINI变体均在5例食管癌中表达下调。结论:双相电泳连接质谱的蛋白质组学方法是快速筛选肿瘤异常表达蛋白的有效手段,HSP27和ANNEXINI的表达失调是食管癌发生过程中的频发事件。

Objective: Proteomics is currently in a phase of technological development and establishment, demonstrating the capacity for high throughput, is a major challenge. It is used in life science research, particulay in the mechanism of tumrogenesis. Yinchenwulin powder, which was extracted from Chinese herbs, is used to treat the patients with atherosclerosis. We investigated the molecular mechanism of Yinchenwulin Powder on the atherosclerotic rats in this paper. Methods: The AS model wistar rats made by feeding with high grease food and AS enplaques were observed in the aorta after one month of feeding. By two-dimension electrophoresis,the proteins were separated, and as well as protein profiles of aorta tissue. Compared with the protein profiles of aorta tissue treated, model proteins in different expression were identified by Image-Master ID Elite 4.01 .Result: There were 1077+68 protein dots in aorta tissue treated,and 1106+84 in aorta tissue. Compared the protein profiles of aorta tissue treated with model, 556 proteins were found as dys-regulated. Among them,196 proteins were observed up-regulated and 364 proteins were seen down-regulated in aorta tissue treated.Conclusion: Yinchenwulin Powder which could treat atherosclerotic rat may associated with those protein regulated by itself,and it will provide a new method of molecular mechanism of Yinchenwulin Powder.

目的:探讨茵陈五苓散对动脉粥样硬化(AS)大鼠作用的分子机制,方法:20只雄性 Wistar大鼠采用高脂饲料喂饲法复制大鼠AS模型,造模1月后抽样检测大鼠主动脉,以发现动脉粥样硬化斑块为造模成功指标。茵陈五苓散治疗 1 个月后分别用固相pH梯度双向凝胶电泳分离茵陈五苓散组和模型组的AS大鼠主动脉组织总蛋白质,银染显色,PDQuest 2 de软件分析差异蛋白质点。结果: 双向电泳图象分析茵陈五苓散组图象的平均蛋白质数为 1077±68,模型组的平均蛋白质数为 1106±84;差异蛋白质有556个,其中有196个表达增强,有360蛋白质表达下降。结论: 茵陈五苓散的抗 AS作用与这些差异表达蛋白质有关,这些差异蛋白质有待质谱分析鉴定,为茵陈五苓散作用的分子机理研究提供了新的思路。
