We previously reported that theintracellular microtubules of normalfibroblasts from foreskin of Chinesechild show specific staining patterns.We here demonstrate several fluores-cent staining patterns of cytoplasmicmicrotubule complex(CMTC)in wellcharacterized tumor cell lines includinghuman stomach cancer cell lines(MGc-80-3 SGC-7901),human esophagealcarcinoma cells(ECa-109),human na-sol pharyngeal carcinoma cells(GNE),human lung cancer cell lines(LTEP-78,LTEP-ai,LTEP-p),HeLa cells,andmouse sarcoma cells(S180-V).Fourmain patterns of GMTC phenotypestermed as FG-,DC-,SO-,and SR-type by Brinkley et al previously innormal and transformed cells are identi-fied in our experiment.The distri-bution of the CMTC phenotypes inpopulations of tumor cells is very dif-ferent from that of normal cells;thefull microtubule complex pattern(FG-type)represents the main CMTC pat-tern of normal cells while the dimini-shed microtubule complex pattern(DC-type)represents that of tumor cells.The results presented here suggest thatthere is a great difference in interphaseCMTC phenotypes between normaland tumor cells.A marked diminutionof microtubular tubulin fluorescencewith special MTOC appearance isshown in tumor cells.Since the ap-pearance of mitotic spindle microtubulesis similar in normal and tumor cellsaccording to our observation,the di-sturbance of interphase CMTC orga-nization is assumed to be the mostimportant problem in the studies ofmalignant transformation.Detailknowledge about the structure andfunction of MTOCs of malignantcells is of interest too.

本实验用管蛋白抗体间接免疫荧光细胞化学方法,观察了我国建株的人胃低分化粘液腺癌MGc 80-3,人胃腺癌SGC-7901,人鼻咽癌上皮样细胞CNE,人食管癌上皮细胞ECa-109,人肺鳞癌LTEP-78,人啼腺癌LTEP-a_1,人肺小细胞癌LTEP-p七株癌细胞和HeLa细胞,小鼠S_(180)-V肉瘤细胞的微管形态。与人的正常包皮成纤维细胞和食管上皮细胞内精细的CMTC结构对比,肿瘤细胞间期的胞质微管普遍有减少或缺如的现象。参考Brin-kley对微管免疫荧光染色图形的分型方法,我们将观察的各种微管染色图形归纳为四种类型,比较各种细胞群体内微管类型的分布。肿瘤细胞群体内多数为微管缺如型和稀疏型,未见典型的丰满型,而正常细胞群体内都是丰满型。同时,肿瘤细胞的MTOC区面积明显增大。分裂期的肿瘤细胞内,有丝分裂器纺锤体微管荧光形态与正常细胞的没有差别。本文对肿瘤细胞间期胞质微管减少和缺如以及MTOC区明显增大的现象及其可能的意义进行了讨论,认为这是癌变机制研究中值得深入探讨的重要课题之一。

Cell hybridization was done by means of polyethylene glycol (PEG)in suspension.Seven kinds of heterokaryons showing pr- emature condensed chromosomes were obtained.A hybridoma cell line had been established which produced monoclonal anti- body anti human complement 4.On the basis of showing prem- ature condensed chromosomes of heterophasic heterokaryons between CHO metaphase cells and bone marrow lymphocytes from patients with leukemia,their potential proliferation index of bone marrow cell population was studied.Here also were discussed some items,the fusioninducing effects of PEG,the mechanism of the increase of leukemia patients'PPI and the subclioning of hybridoma cell line etc.

本文报告了1980年以来我们在细胞杂交技术及其应用研究中取得的各项阶段性成果。在细胞杂交技术方面,用分子量在1000、2000、4000、6000的PEG为融合介质,获得了中国仓鼠成纤维细胞Wg3-h、人HeLa细胞、人胃腺癌细胞SGC-T901的同种异核体,以及雄性麂肺细胞KIZ-7901-中国仓鼠成纤维细胞Wg3-h、中国仓鼠卵巢细胞CHO-人外周血港巴细胞、中国仓鼠卵巢细胞CHO-正常人骨髓细胞、中国仓鼠卵巢细胞CHO-白血病患者骨髓细胞的异种异核体,并成功地显示了上述各种细胞组合的PCC图象。获得了在体外长期传代培养的小鼠骨髓瘤细胞NS-1-Ag8-BALB/c小鼠脾脏淋巴细胞以及小鼠骨髓瘤细胞SP2/0-Ag14-免疫后的BALB/c小鼠脾脏淋巴细胞的种内杂种细胞系。在细胞杂交技术的应用方面,重点在诱导“熟前凝聚染色体”(PCC),分析白血病患者骨髓细胞的群体增殖潜能。并据以判断白血病患者病情进展、疗效和预后,以及制备分泌抗人C4单克隆抗体的杂交瘤细胞系。本文对PEG的融合性能、白血病患者PPI值升高的机制、杂交瘤细胞的亚克隆化等问题进行了初步探讨。
