Objective To study the cell cyclical characteristics of human gastric cancer line cultured in vitro treated by extracts of Allium Fistulosum.L (AF).Methods After MGC 803 was treated with extracts of AF,the ultrastructure of MGC 803 was observed by electron microscope, and cell DNA contents, cell cycle distribution, apoptosis and differentiation of MGC 803 were detected by flow cytometry.Results (1) Under transmission electron microscope,MGC 803 cell membrane kept completed, cell ultrastructure changed with typical apoptosis and differentiation.(2) The results of flow cytometry showed that① After being treated with 1.25% AF, apoptotic rates of MGC 803 were 0.6%,1.6%,6.1%,13.5% respectively,diploid rates were 0%,0.3%,35.8%,93.9% respectively at 24 h,48 h,72 h,96 h.②At 24h,apoptotic rates of different groups (1.25%,2.5%,5.0%,10.0%) treated with extracts of AF were 0.61%,4.1%, 6.2%, 24.9% respectively,diploid rates were 0%, 10.6%, 34.7%,72.2% respectively.Low concentration of green onion induced differentiation of MGC 803 cells mainly,but high concentration of green onion induced both differentiation and apoptosis of MGC 803 cells simultaneously. Effects of induction of differentiation and apoptosis depended on exposed time and concentration of green onion. With the concentration and time increased, diploid rate and apoptotic rate went up, while aneuploid rate decreased.A typical subdiploid peak in front of G0/G1 phase was observed by flow cytometry. Apoptosis of MGC 803 cells occurred in G0/G1 and S and G2/M phases, without cyclical specificity.Conclusion The extracts of AF.L could inhibit gastric cancer cell's growth and proliferation, induce both differentiation and apoptosis of gastric cancer cell line and its anti-tumor effect has no cyclical specialty.

目的观察大葱提取液抗胃癌作用的细胞周期特点。方法将大葱提取液作用于体外培养的人胃癌细胞MGC803后,采用电镜观察MGC803细胞超微结构变化;应用流式细胞仪分析MGC803细胞DNA含量及周期分布,检测其分化与凋亡情况。结果(1)透射电镜下观察:大葱作用后的癌细胞细胞膜完整,出现分化和凋亡的形态学变化。(2)流式细胞仪检测:1.25%大葱提取液作用MGC803细胞24h、48h、72h、96h,凋亡率分别为0.6%、1.6%、6.1%、13.5%,二倍体比例分别为0%、0.3%、35.8%、93.9%;1.25%、2.5%、5.0%、10.0%大葱提取液作用24h,凋亡率分别为0.6%、4.1%、6.2%、24.9%,二倍体比例分别为0%、10.6%、34.7%、72.2%。低浓度组以诱导分化作用为主,高浓度组同时可诱导细胞凋亡,呈剂量和时间依赖性。实验组细胞随大葱液浓度和作用时间增加,异倍体细胞比例逐渐减少,细胞凋亡率逐渐增加。DNA含量出现二倍体峰和亚二倍体峰~凋亡峰。大葱提取液抑制胃癌细胞可发生在G0/G1期、S期和G2/M期,无周期特异性。结论大葱提取液能明显抑制体外培养的胃癌细胞生长增殖,既可诱导...
