Objective: To analyse the effect of leucocytes suspension infusion in chemotherpy of cancer patients and to study the white blood cell maintenance in patients' blood. Methods: The blood was collected from donors whose blood types were same with the recipients' and whose physical examination and chemical tests were all normal, then the leucocytes were isolated from the blood and centrifuged twice being prepased for concentrated 30~45ml plasma suspension. The suspension (1.3~2.2×1010 cell each time) was used to treat 20 cases of cancer patients for 3~5 times (whose white blood cells were less than 1.7×109/L and accompanied with infection. Forty-eight hours and one week after infusion, the white blood cells of the patients were counted and temperature and infection condition were observed. Resultes: Forty-eight hours after leucocytes infusion, the WBC count of 20 patients with chemotherapy were higher than before (from 1.13×109/L raise to 5.03×109/L), and in whom 17 patients (85%) had the WBC count up to 2.3×109/L and only 3 cases kept unchanged. The effective rate was 85% and 65% of them was more significant. Conclusion: The cancer patients treated with chemotherapy usually have leucopenia and accompanied with infection, though treated with blood transfusion,antibiotics or leucocyte-increasing drugs, the therapeutic effect was not ideal. Supplementary leucocyte suspension infusion would get good curative effect for these patients.

目的 :了解白细胞悬液在癌症患者化疗中应用的效果与患者体内白细胞维持时间。方法 :采集与受血者血型相同 ,体检及化验检查均合格的献血者血液分离白细胞 ,经2次离心法浓缩成30~45ml血浆悬液 ,对在化疗中致使白细胞计数低于1.7×109/L ,伴有感染的20例癌症患者输注白细胞悬液(1.3~2.2)×1010 个/次 ,3~5次 ,观察输注后48小时和1周时白细胞计数、体温及感染控制情况。结果 :20例癌症化疗患者输注白细胞悬液48小时后 ,白细胞计数由输注前的平均1.13×109/L升至输注后的5.03×109/L,其中白细胞计数升高2.3×109/L以上者17例(85 %) ,白细胞计数无明显升高或降低者3例(15 %)。用此法治疗后85%有效 ,其中65 %效果尤为明显。结论 :对癌症患者在化疗中致使白细胞数减少 ,伴有感染者 ,经输全血、用升白细胞药物及抗感染治疗48小时无明显效果时 ,加输白细胞悬液3~5次有明显疗效。

Objective:To investigate the function and expression of heat shock protein70in gastric cancer cells induced by high temperature and Newcastle Disease Virus infection solely or associated with each other in vitro.Methods:At first,by high temperature and infection with NDV in vitro to probe into the alteration of morphology and proliferation of BGC-823gastric cancer cells by means of light microscopy and MTT assay.The apoptosis,alteration of every phase of cell cycle for BGC-823gastric cancer cell line and the overexpression of HSP under high temperature and NDV was measured by using fluorescence activated cell sorter(FACS)and RNA dot blot.Results:The function of killing by high temperature and NDV infection was shown.The overexpression of HSP70and its mRNA was detected in the cells under various stresses and induced the apoptosis of cancer cell,reduced S-phase and G2-phase cells,mean-while,proliferation idex(PI)declined.Compared with negative control group,the effect of various stresses was significant (P<0.05).Conclusions:High temperature and NDV used solely or asso-ciate with each other could lead to overexpression of HSP70and had antitumor activity in vitro.

目的:研究加热法及新城疫病毒(NDV)单独或联合体外诱导BGC-823胃癌细胞热休克蛋白70(HSP70)的表达及其意义。方法:通过加热及NDV感染BGC-823胃癌细胞,经倒置显微镜观察细胞形态变化,噻唑蓝(MTT)比色法检测胃癌细胞的增殖状况,经流式细胞仪分析法及RNA斑点杂交检测感染及加热后细胞凋亡、细胞分裂周期各时相的变化及胃癌细胞HSP70的表达。结果:加热及NDV感染对BGC-823胃癌细胞的杀伤作用明显。加热和感染均可使细胞高表达HSP70及其mRNA,细胞凋亡率增加,G2、S期细胞减少,增殖指数(PI)降低,与阴性对照组比较有显著性差异(P<0.05)。结论:加热法与NDV感染法单独或联合应用均可增强胃癌细胞HSP70及其mRNA的表达且有较强的杀瘤作用。
