The adherence of S.mutans, which is known to be caries-conducive in experimental animals, toextracted teeth, artificial teeth,nichrome steel wires, beads ofhydroxyapatite and other objects inthe presence of sucrose has beendemonstrated by a number of investi-gators. But adherence of S. mutans toslides has not yet been observed undermicroscope. We inoculated seven S.mutans serotypes a, b, c, d, e, f,and g, respectively into test tubes,each containing a small slide and 5ml. of trypticase peptone water supple-mented with 10 % sucrose (W/V).They were then incubated at 37℃ inan atmosphere of N_2 95 % and CO_25 % for 24-72 hours. Five controlbacterial cultures containing staph.aureus, β-haemolytic streptococci,α-haemolytic streptococci, E. coli, andS. typhi respectively, were incubatedin the same conditions as the S.mutans except that free oxygen is pre-sent in the control cultures.To exam-ine microscopically, the small slideswere rinsed with tap water to removethe unadhered bacterial cells andstained by Gram's method. The results indicated that all theseven serotype strains of S. mutansappeared in numerous clusters adheringto the slides, while there were notany bacterial clusters on the slides inthe five control cultures. The S. mu-tans bacterial clusters adhering toslides might be divided into two formsaccording to their appearance: thelarger and looser form and the smallerand denser form. The strain OMZ 176(d type) always appeared in smaller,denser and isolated clusters, whichmight be a specific appearance usefulfor identification. The method of smallslide culture, owing to its simplicity,could be applied to the serotyping offluorescent antibody stain and to theisolation and identification of S. mu-tans from the dental plague samples.The mechanism of adherence of S.mutans and the appearance of d typebacterial clusters were discussedbriefly.
