Objective: We examined the prevention and cure effect of the Alcohol extract of spica prunellae (AEP) on experimental diabetic nephropathy in rat. Objective: We examined the prevention and cure effect of the Alcohol extract of spica prunellae (AEP) on experimental diabetic nephropathy in rat. Methods: The AEP was obtained by extracting spica prunellae with ethanol and chloroform. After 4 weeks experimental diabetic model was induced by i.p. rats streptozotocin(STZ), DM rats were administratred the AEP 100mg·kg -1 ,200mg·kg -1 for 4 weeks, blood glucose, kidney function and activity of kidney aldose reductase were determined. Results: The AEP decreased significantly urinary protien in diabetic rats. The AEP 100mg·kg -1 decreased blood urea nitrogen(BUN) and creatinine(Cr) in diabetic rats. The AEP deceased significantly activity of kidney aldose reductase in diabetic rats. Conclusions: The AEP had certain degree of preventing diabetic nephropathy. The results suggested that the AEP defect diabetic nephropathy by inhibiting activity of aldose reductase to decrease associated tissue or organ injuries caused by accumulation of sorbitol.

目的 :观察夏枯草醇提物对实验性糖尿病大鼠肾脏病变的防治作用。方法 :夏枯草经乙醇、氯仿提取得醇提物 (AEP)。腹腔注射链脲佐菌素后 4周 ,造成大鼠糖尿病模型 (DM ) ,灌胃AEP 10 0mg/kg、2 0 0mg/kg 4周 ,测定血糖、肾功能和肾组织醛糖还原酶活性。结果 :AEP能显著降低糖尿病大鼠尿蛋白 ,AEP 10 0mg/kg能降低血清尿素氮、肌酐含量 ;能显著降低DM大鼠肾组织醛糖还原酶 (AR)活性。结论 :AEP对糖尿病大鼠并发肾脏病变有一定的防治作用 ,其作用机制可能通过抑制醛糖还原酶活性以减轻山梨醇蓄积造成相关组织器官的损害。

Objective To investigate the effect of Fosinopril,which has compensatory feature of dual elimination,on preventing and treating diabetic nephropathy in rats.Methods Rats were randomly divided into normal control group (C),streptozotocin diabetic group (D),diabetic rats treated with fosinopril [8mg/(kg·d)] group (M).Kidney weight/body weight,urinary volum,creatinine clearance rate (Ccr),urinary albumin and β 2-MG,urinary and serum ET-1 and TNF-α were measured at first,second and fourth week after treatment,the glomerular parameters were studied at the fouth week by using computer morphometric image analysis.Results Comparing with group C,kidney weight/body weight,urinary volum,Ccr,urinary albumin and β 2-MG in group D were increased significantly.The level of ET-1 in urine increased by 14 folds and by 0.2 folds in serum.Its elevation in urine was prior to in serum.TNF-α in urine increased also markedly but there was no change in serum.Significant decreasing in all above parameters were observed in group M compared with group D.Kidney morphometric image analysis showed that matrix and basement membrane area density decreased from 0.50±0.80 of group D to 0.25±0.02 of group M while capillary tuft area density increased from 0.50±0.08 to 0.77±0.02.Conclusion Fosinopril can significantly modify diabetic renal injury produced by glomerular hypertension and hyperfiltration,reduce urinary protein excretion,inhibit production of ET-1 and TNF-α in kidney and inhibit glomerular hypertrophies and mesangial matrix expansion.Fosinopril is very effective in preventing and treating diabetic nephropathy.

目的 探讨福辛普利对糖尿病鼠肾病的防治及机理。方法 设正常组 (C)、糖尿病组 (D)和福辛普利组 (M) ,检测有关指标及肾病理。结果 D组尿蛋白、内皮素 - 1等均较 C组明显升高 ,M组上述指标均较 D组下降 ,病理示系膜基质减少。结论 福辛普利能缓解糖尿病鼠肾高滤过 ,降低尿蛋白、抑制肾炎症细胞因子 ,从而防治肾小球硬化

Objective To assess the effect of rhein on diabetic nephropathy in rats with type 2 diabetes mellitus. Methods Diabetes was induced by high-lipids and high-sucrose diets (20% sucrose, 10% lard, 2.5% cholesterol, 1% cholic acid and 66.5 % regular chow, w/w) for 1 month and then intraperitoneal injection with 25 mg/kg streptozoticin(STZ) in female inbred Wistar rats. Blood and urinary biochemistry and renal morphology were evaluated at divers times. Results Rhein ameliorated hypertriglyceridemia and hypercholesterolemia, but had few effect on the level of plasma glucose in both rhein-prevented group and rhein-treated group. Excretion of urinary protein reduced markedly and stable-state plasma glucose (SSPG) level decreased in both groups as compared with diabetic group. Examination of kidney sections from both rhein-prevented group and rhein-treated group showed that the structural lesions of glomeruli including mesangial expansion, diffuse glomerulosclersis and thickening of glomerular basement membrane, as well as artery injury such as arteriolar hyalinosis, atherosclerosis were markedly improved as compared with diabetic group. Morphometry of glomeruli from both rhein-prevented group and rhein-treated group showed that glomerular grow and mesangial hypertrophy occurred in diabetic group were ameliorated. The accumulation of fibronectin examined by immunostaining in glomeruli was reduced in both groups. Conclusion Therapeutic intervention with rhein can halt the progression of diabetic nephropathy and prevent the development of glomerulosclerosis and vascular injuries in this animal model of type 2 diabetes mellitus.

目的 观察大黄酸防治2型糖尿病肾病的作用。方法 以高糖高脂饮食联合低剂量链脲佐菌素(STZ)注射方法诱导出2型糖尿病肾病大鼠模型后,分模型组、预防组和治疗组。后2组分别在STZ注射后1周和1个月后给予大黄酸100 mg/kg剂量灌胃,每日1次,连续6个月,在不同时间进行形态学和有关生化指标的观察。结果 大黄酸预防给药和治疗后的观察结果显示,大黄酸明显降低糖尿病大鼠的24 h尿蛋白排泄,改善肾重指数。肾脏组织病理学显示,大黄酸处理后的糖尿病大鼠肾小球和丝球体的面积明显减小,襻腔扩张减轻,系膜增生和细胞外基质减少。大鼠肾小球纤连蛋白沉积明显减弱。大黄酸也明显降低糖尿病大鼠的血脂水平。以上作用随着给药时间的延长,效果越来越明显。胰岛素抑制试验表明,在大黄酸预防给药或治疗6个月后,糖尿病大鼠的血浆稳态葡萄糖水平(SSPG)显著降低。以上结果比较,以预防组效果为佳。结论 大黄酸通过降低糖尿病大鼠的尿蛋白排泄、减轻肾脏肥大、改善胰岛素敏感性、降低血脂水平,有效地防治2型糖尿病肾病。
