This essay presents 2 cases,one suffering from chronic granulocytic leukemia,the other from malignant lymphoid tumor, who received autologous bone marrow transplant (ABMT) with purgatation through combined hyperthermia and interferon, with the marrow temperature kept at 43℃ for 70 minutes. The hematopoiesis of the marrow was restored normal without delay after the treatment. The result shows that the combined purgatation is both effective and simple to perform.

本文采用自体骨髓体外高热联合干扰素净化方法.对1例慢性粒细胞白血病及1例恶性淋巴瘤病人行自体骨髓移植术。每毫升骨髓干扰素含量300U,骨髓处理温度控制在43℃,70min。水后骨髓造血未见延迟恢复。一例病人已恢复工作1.5年。提示该净化方法简单、有效.可在基层单位推广。

To assess the potential of photochemotherapy for the in vitro purging of residual leukemia cells from autologous bone marrow transplants ,we studied the effect of YHPD combined with light irradiation on human leukemia cell line K 562 and normal human hematopoietic progenitor cells by using the methods of trypan blue exclusion test and colony assay. To assess the potential of photochemotherapy for the in vitro purging of residual leukemia cells from autologous bone marrow transplants ,we studied the effect of YHPD combined with light irradiation on human leukemia cell line K 562 and normal human hematopoietic progenitor cells by using the methods of trypan blue exclusion test and colony assay. We also investigated the influence of YHPD dose and light exposed time on this photosensitivity of K 562 cells. It was found that YHPD or light irradiation alone had no apparent cytotoxicity while the application of YHPD plus light exposure showed the photosensitization. Decrease in cell viability was linear when plotted on a logarithmic scale from 0 to 5 min (light irradiation) or 0 to 5 mg/L(YHPD). YHPD plus light irradiation was selectively more efficient for killing K 562 cells than normal human marrow progenitors. Under the condition of a 5log reduction in the survival of K 562 cells, 15.97% of normal human CFU GM survived. These results indicate that the photochemotherapy mediated by YHPD might be used for purging bone marrow in vitro.

为研究光卟啉(YHPD)加光照对白血病细胞株K562的杀伤作用及对正常造血祖细胞的影响,通过细胞体外液体培养活细胞计数、体外半固体培养集落测定,以探讨该法用于体外净化自体移植骨髓中残留的白血病细胞的可能性。观察不同光敏剂剂量和光照时间的变化对杀伤白血病细胞作用的影响,结果表明,单用YHPD或单独日光荧光照射对K562细胞和正常人造血祖细胞均无明显杀伤作用。二者联合使用表现光敏效应。细胞存活率的对数与YHPD剂量、光照时间呈线性相关。YHPD加光照对K562细胞具有强大杀伤作用,对正常造血祖细胞影响较小,在能够杀伤白血病细胞大于5个对数级,其体外已不能形成集落的情况下,正常人骨髓-巨噬细胞集落形成单位(CFU-GM)尚有15.97%,提示该法可望用于骨髓体外净化

Objective[WT5BZ]:To compare the speeds of engraftment and concerning clinical data between autologous peripheral blood stem cell plus bone marrow transplants (APBSCT+ABMT)and simple autologous bone marrow transplant(ABMT).[WT5HZ]Methods:[WT5BZ]APBSCT+ABMT( n =10) and ABMT( n =10) were used on the treatment of acute leukemia and lymphoma patients,their data were studied.[WT5HZ]Results:[WT5BZ]The average days reach WBC>1.0×10 9·L 1 ,N>0.5×10 9·L 1 and Plt>50×10 9·L 1 were 10,10.5,12.5 in APBSCT+ABMT group and 15,17,24 in ABMT group respectively( P <0.05).There was a significant decrease in the number of plasma and platelets transfusions in the APBSCT+ABMT group as compared with ABMT group( P <0.01).The rate of Tienam use was significantly greater in ABMT group than in APBSCT+ABMT group( P <0.05, P <0.01).There was a significant difference about the rate and degree of bleeding between these two groups ( P <0.05),and also a significant shorting of living in laminar air flow bioclean room in APBSCT+ABMT group than in ABMT group ( P <0.01).No difference was seen between two groups about reducing of hemoglobin and the number of red blood cells transfusion( P >0.05).[WT5HZ]Conclusion:[WT5BZ]APBSCT+ABMT can improve the rate of hematologic recovery,and thus,decrease the rate of infection,bleeding,transfusion,antibiotics usage and length of hospitalization,it is an effective therapy for malignancies.
