Objective To study the relation that myocardial Caspase-3 mRNA with activity changes and medicine influence in regions after permanent ischemia and ischemia reperfusion.Methods Build up the domestic rabbit model of myocardial ischemia reperfusion.A total of 17 rabbits were divided into 4 groups:ischemia group,IR group,IR+AC-DEVD-CHO group,and sham operation group.The ischemic model were observed at 1.5h after ischemia followed by reperfusion at 2,4,8,12,24 and 48h.Adopt the fluorescence analysis method to examine caspase-3 activities.The activity of Caspase-3,the apoptotic cardiomyocytes and the myocardial infarction area were determined by fluorescent assay,TUNEL method and TTC dyeing,respectively.Results ①Ischemia group and IR group compared,ordering in each time,the Caspase-3 of the latter is obviously up-adjusted,the boundary area of apoptotic cell enlarging,the infarction area extension,particularly at 24h,the difference significant,P<0.01.②IR group and IR+AC-DEVD-CHO group compared,the obstruct of Caspase-3 activity at each time in latter,the apoptotic cell reduce,and the infarction area contract,particularly at 24h,P<0.01. ③In ischemia!group,IR group,and IR+AC-DEVD-CHO group,there present positive relativity at the Caspase-3 activity,the count of apoptotic cells and the myocardial infarction area.Conclusion ①The boundary area of Ischemia reperfusion activate the activity of Caspase-3,make the apoptotic cell increase,the infarction area enlarge.The ischemia group influences larger to the myocardial.Stop the activity of Caspase-3 can make apoptotic cell number descend,the infarction area contract.②The boundary area Caspase-3 activity and the apoptotic cell quantities affect the occurrence,development and structure reconstructions of the myocardial infarction,and provide a time window and new strategy for curing the myocardial infarction.

[Objective] To check and compare the different chinese herbal medicine influence on ALI through their function against oxy-radicals.[Methods] Divided the laboratory animals into 10 groups randomly,as these(the methods,injection,of the treatment to each member below were the same,here mentioned the different reagents for concision):Group A1,6 hrs after oleic acid + NS;Group A2,8 hrs after NS only;Group B1,6 hrs after oleic acid + VitC injection(VC);Group B2,8 hrs after VC;Group C1,6 hrs after oleic acid + Shenmai Injection;Group C2,8 hrs after oleic acid + Shenmai Injection;Group D1,6 hrs after oleic acid + Daidzein(Puerarin Injection);Group D2,8 hrs after oleic acid + Daidzein(Puerarin Injection);Group E1,6 hrs after oleic acid + Qingkailing Injection;Group E2,8 hrs after oleic acid + Qingkailing Injection.Evaluated the level of free oxygen radical by detect MDA,T-SOD,GSH,and GSH-Px;evaluated the degree of acute inflammation and edema(lung)by lung coefficient;mention about the degree of the inflammation,it had to take the resort to the examination under light microscope pathologically,used the tissue from apex of right lung.[Results] ①Lung coefficient:no obvious differences among the "6 hrs Groups"(A1,B1,C1,D1,E1)compared with the "8 hrs groups"(A2,B2,C2,D2,E2),and in the later groups,Group E2 was remarkable higher than Group A2;② MDA:Group C1 increased mostly among "6 hrs groups",and so was Group C2 among "8 hrs groups";③ T-SOD:no marked differences among the "6 hrs Groups",and in "8 hrs groups",Group E2 was lower than Group C2 obviously;④ GSH:Group D1 and C2 were the highest in "6 hrs groups" and "8 hrs groups" respectively;⑤ GSH-Px:no changing had been observed;⑥ the degree of congestion and edema(pulmonary alveoli):Group A1,B1,C1,D1,E1 were alike,and Group A2>D2>C2>B2≌E2.[Conclusion] VC,Shenmai Injection,Daidzein(Puerarin Injection),and Qingkailing Injection,all are available anti-oxyradical medicine.Some diversities of them four are:ⅰ Shenmai Injection is weaker than others in lowering MDA;ⅱ when step up T-SOD,Qingkailing Injection is almost faint;ⅲ Daidzein(Puerarin Injection)and Shenmai Injection act a little stronger for GSH increase.The observation,6 hrs after injury,are resemble on the whole;but if injured for 8 hrs,the groups treated with VC and Qingkailing Injection are milder than with Shenmai Injection and Daidzein(Puerarin Injection).
