Using standard intracellular recording and staining techniques,4 chandelier interneurons were sampled and anatomically confirmed from 39 physiologically identified interneurons in rat hippocampal slices.Compared to pyramidal and granule cells,all 4 chandelier interneurons(CAI region, n=3,dentate gyms,n=1)had typical action potentials of interneurons,i.e.,a shorter duration of action potential was followed by a deep after hyperpolarization.However,all these 4 cells exhibited a varying degree of spike frequency adaptation in response to depolarizing current injection, which usually do not occur in basket interneurons.Morphologically,a-conal terminals of chandelier cells were beaded and exclusively distributed in the stratum oriens of the CAI and the granule cell layer of the dentate gyms,where the initial segments of pyramidal and granule cells are located.These results indicate that the chandelier interneurons differ from basket interneurons in the rat hippocampus with in physiology and in morphology.

We have investigated the changes of calcium-activated neutral proteinases (calpains) activity during reperfusion after transient cerebral ischemia by using the four-vessel occlusion model of ischemia in rat. The results indicated that calpain I and calpain I activities increased significantly and neuronal density of the CAI region of the hippocampus decreased correspondently. It is concluded that calpains may be responsible for ischemic damage to the brain.

本文采用大鼠全脑缺血模型,观测脑缺血再灌流脑组织Ca~(2+)依赖性中性蛋白酶(calci-um-activated neutral proteinase,calpain)活性的变化及海马C_(A1)区神经元损害改变。结果显示脑缺血再灌流脑组织calpain Ⅰ和calpain Ⅱ活性都明显升高(P<0.01),C_(A1)区神经元密度相应下降,提示calpains在脑缺血损害过程中可能起一定作用。

To investigate the reperfusion neuronal damage and heat shock protein(HSP) 70 expression in incomplete cerebral ischemic mice, the microstructure of forebrain was observed by TUNEL assay as well as HSP70 immunohistological method 24 to 48 hours later after temporarily occluding animal's bilateral carotids. It was found that extensive neuronal shrinkage with irregularly condensed nuclear appeared in the pyramidal layers of hippocampal CAI region 24 hours later after ischemia. Both the condensed chromatin abutting the nuclear membrane and the ultrastructure alterations such as mitochonderial swelling could be seen in this area. There existed abnormal morphological features in the II~VI layers of the cerebral cortex, cinguli and dentatus gyms, candoputamen and hippocampal CA3 regions to varying extent, and the cellular density in lateralis habenularis was greatly decreased. At the same time a great number of TUNEL-positive cells were scattered in the cortexes and hippocampus. The positive HSP70--immunoactive cells were found in the above regions of forebrain, while strong positive immunostain lay in the pyramidal cells in hippocampal CA3 and II--VI layer of the cerebral cortex. These results indicate that both necrosis and apoptosis may relate to the reperfusive forebrain damage in mice, and the HSP70 expression also involve in the forebrain's responses to cerebral ischemia-reperfusion stress.

目的:研究小鼠不完全脑缺血再灌期损伤的特点及热休克蛋白(HSP)70的表达。方法:暂时阻断小鼠双侧颈总动脉,再灌后24或48小时取材作光镜、电镜、TUNEL染色及HSP70免疫细胞化学观察。结果:再灌24小时海马CAI区见大量核固缩的锥体细胞,电镜下可见核染色质聚积于核膜边和线粒体肿胀或空泡化等改变。皮层Ⅱ~Ⅵ层、扣带回、齿状回、尾壳核、海马CA3等处也见程度不同的受损细胞,缰外侧核细胞明显减少。大量的DNA原位末端缺刻标记(TUNEL)阳性细胞呈片状或散在出现于再灌24小时的皮层和海马。HSP70免疫阳性细胞可见于上述前脑各区,但强的 HSP70免疫阳性常现于海马CA3区和皮层Ⅱ~Ⅵ层细胞内。结论:坏死和凋亡均可见于小鼠缺血再灌性前脑损伤,并有HSP70的表达增强。
