Recent studies on the immunogenicity of mammalian ovaries have shown that heteroim-munization with ovarian tissue will induce antibodies with which several ovary antigenscan be detected.Through various biochemical and immunological studies,it has been shownthat these antigens are located on the surface of the ovum,i.e.zona pellucida.Moreover,it has been reported that this surface antigen-induced antibody could block sperm penetra-tion of the zona pellucida in vitro and inhibit fertilization in vivo.It appears that the rabbit anti-zona antiserum is not speices-specific so far as its ability to inhibit fertilization invivo and in vitro is concerned.More recently,Shivers and Stevens have shown that active immunization with porcinezona in dogs,marmosets and baboons is successful in blocking fertilization.Presently,scientists in this area are isolating pure zona antigens and pursuing intensive studies with thefinal goal of going to human trials.Although the biochemical,physiological and immunological studies on mammalianzona pellucida are well documented,but to develop a vaccine for large scale applicationneeds further reproductive immunological studies on hetero-zona-antigens from other non-mammal animals with rich availability.Based on this background,the present investigationhas been undertaken to study the zona pellucida of four fresh water fishes and their im-munological properties,namely,Cyprinus carpio,Ctenopharyngodon idellus,Anistichthysnobilis and Hypophthalmichthys militrix.Ovulation was induced artificially in different animals by the injection of gonadotropichormone.Some specimens were collected from the ovaries of the laparotomized females.After its isolation,the zona pellucida(ZP)was kept frozon in the refrigerater until use.200ZP specimens were thawed and homogenized;one c.c.homogenate was emulsified with anequal volume of Freund's complete adjuvant(FCA)and 2 c.c. of this emulsion were injectedsubcutaneously into the scapular region of rabbits,a booster injection in Freund's incompleteadjuvant being given 2 wks.or one month later.Some animals were given 7-10 successiveinjections.After the final injection,the rabbits were bled by carotid cannulation or heartpuncture for the preparation of serum.Serum samples were analysed on polyacrylamide gel electrophoresis.Titration ofantiserum against different ZP coated sheep or rabbit tanned RBC was done by the passivehaemagglutinin test.The classic slide precipitation test and agar gel plate double diffusiontechnique were used to detect precipitation antibodies.100 ZP specimens were homogenized and centrifuged at 2200xg for 15 min.,the super-natant being subjected to polyacrylamideb gel electrophoretic analysis.A radioisotope I~(131)labelled ZP was prepared for the specific antigen-antibody reaction test.Hetero-antigenicity of fish ZP:Polyacrylamide gel electrophoretic analysis showedthat fish ZP had the similar pattern to rat ZP and bacterial mucopotysaccharide.With pro-tein dye,amido black 10B,a single blue black band could be stained.Passive haemagglu-tination titre of the rabbit anti-fish ZP antiserum against different ZP antigens could reach2~(10).The precipitin bands formed between antiserum and ZP antigens are presented in thereport.The results revealed that many precipitin bands were formed between differentspecies.It was very interesting to note that a dim precipitin band sometimes could befound in large proportions of rabbit anti-fish ZP antiserum and rat ZP antigen.Similarresults were also observed on the slide precipitation test.Analysis of the rabbit anti-fish ZP antiserum:During the active immunization period,serum sample analysis revealed that the γ globulin band was gradually thickened.Whenthe antiserum was incubated with the I~(131) labelled ZP for several minutes before electrop-horetic separation,the resultant γ globulin band possessed a high radioactivity count.The active immunization with fish ZP antigen and passive immunization with rabbitanti-fish ZP antiserum on fertilization in vivo are in progress in our laboratory and will bepublished in a separate paper.From the above results,we believe that the composition of ZP of different animals innature might be chemically and immunologically related.We have found that rabbit anti-fish ZP antiserum is not species-specific so far as the in-vitro cross-reactivity is con-cerned.

从淡水鱼类(鲤、草、鲢、鳙)卵中显微分离出的透明带,经聚丙烯酰胺凝胶圆盘电泳,对蛋白染料氨基黑10B 着染,呈单条蓝黑色带,它和大白鼠卵透明带、细菌粘多糖及唾液粘蛋白有类似的电泳谱及迁移率。用鱼卵透明带经福氏佐剂乳化后免疫家兔,可诱发家兔产生抗鱼卵透明带抗体。被动血凝试验测得抗体滴度为1000,玻片沉淀反应及琼脂凝胶平板双扩散试验表明,此抗血清可以和不同种鱼透明带及大白鼠透明带起交叉反应,猴抗鱼透明带抗血清可以和猪卵透明带起交叉反应。鱼卵透明带可以成功地用放射性同位素~(131)Ⅰ进行标记。用鱼卵透明带主动免疫家兔所诱发抗血清的电泳分析表明,丙种球蛋白区带随免疫时间加长而逐渐加深加宽.~(131)Ⅰ标记的鱼透明带与兔抗鱼卵透明带的抗血清一起混合后再电泳,丙种球蛋白区带的放射性计数率最高。初步认为鱼卵透明带的化学成分及免疫学特异性可能与哺乳动物存在着一定关系,有一定的共同抗原性,这为寻找丰富的异种卵透明带抗原提供了一定的线索。
