To compare the sensitivity of DNA probe in detection of Plasmodium falciparum, three different DNA probes were prepared. P. falciparum genomic DNA was prepared from cultured parasite, pBF_(13) DNA was produced from the clone which was selected from P. falciparum genomic DNA library containing single copy gene, and pBF_4 DNA was produced from the clone containing highly repeated gene sequence. To compare the sensitivity of DNA probe in detection of Plasmodium falciparum, three different DNA probes were prepared. P. falciparum genomic DNA was prepared from cultured parasite, pBF_(13) DNA was produced from the clone which was selected from P. falciparum genomic DNA library containing single copy gene, and pBF_4 DNA was produced from the clone containing highly repeated gene sequence. The 3 kinds of DNA probes were labelled with~(32)P and hybridized with P. falciparum DNA on nitrocellulose paper at the same condition. The results showed that the sensitivity of pBF_(13) probe was 10 times less than the whole genomic DNA probe and pBF_4 DNA probe was 10 times more sensitive than the whole genomic DNA probe. The whole genomic DNA probe could detect 100pg DNA of the parasite.

The gene encoding pro-ST of heat-stable enterotoxin(ST) of enterotoxigenic Escherichia coli(ETEC)was fused to the gene encoding the B subunit of heat-Labile enterotoxin(LT-B). And based on this clone, a series of plasmids harbouring various tandem repeat of ST gene from 2 to 5 copies fused to LT-B fusion gene showed the influence bewteen ST and LT-B. The ELISA titers of ST raised as the ST tandem repeat gene copies increased. On the other hand, the ELISA titers of LT detected were going down. This study demonstrated that ST antigenicity could be raised by increasing the ST tandem copies of the clone, and provided a significant experience for the study of ETEC recombinant polyvalent vaccine.

The vaccine expressing plasmid was developed in which the tandem repeat genes coding for two copies of the sequences of 141 160 amino acid residues and 21 40 amino acid residues of VP1 protein of FMDV were fused to a carrier gene coding for β galactosidace. The vaccine can be expressed in E. coli. Animal experiment result showed that this vaccine elicited high levels of neutralizing antibody and specific T cell proliferation in guinea pig and protected guinea pigs against type O FMDV injection.

用FMDV疫苗免疫接种是预防口蹄疫的主要手段之一 .研制既含B细胞表位又含T细胞表位的基因工程疫苗对口蹄疫的防治具有更强的保护能力 .用化学方法合成O型口蹄疫病毒 (FMDV)外壳蛋白VP1中 2 1~ 40位肽段的T细胞表位基因 ,与 1 41~ 1 60肽段的B细胞表位基因串联后 ,与 β 半乳糖苷酶基因相连 ,构建成一重组质粒 ,并在大肠杆菌中表达出融合蛋白疫苗 .动物实验表明 ,该疫苗不仅能诱导豚鼠产生滴度很高的中和抗体 ,而且产生大量对病毒专一性T细胞 ,与不含 2 1~ 40肽段的疫苗相比 ,用该疫苗免疫的豚鼠血单个核细胞对FMDV的反应性提高 7倍以上 ,并能抵抗病毒攻击 .说明该疫苗可同时激活体液免疫及细胞免疫反应 ,有较强的保护作用 .
