When using spectrophotometry to determine the minute quantity et cobaltus 〔Ⅱ〕ion in BI water we used NH 4SCN as color agent and saturated NaF solution as cover aget and acetone as stabilizer in the solution with Ph value of 5.0~7.0.We can directly determine the adsorption at 618 nm and with deionized water as reference and then determine the absorption at the standard sample to make work curve.Then we determind the absorption of the sample and obtained the content of cobaltous 〔Ⅱ〕 ion through the work curve.By this method,the recovery was among 98.5%~100.5%.Compared with chemical method ,the spetrophotometny′s relative standard deviation CV was 5.6‰,while the chemical method′s standard deviation CV was 25.8‰ it′s accuracy and property are both good,its appearance was good and with betteor selection.The disturbance at this method was small and its operation was convenient.There is no need to handle the sample and it′s determination was repid.It was adaptable to production in the chemical industry and the detection of enviroment.

Uricase from Candida utilis was isolated by bi-water phase extraction, then through the separation procedures including DEAE-Sepharose FF, Phenyl Sepharose CL-4B and Xanthine-agarose chromatographies, uricase was purified to electrophoretic homogeneity from liquid cultured cells of the Candida utilis. the yield of enzyme activity was 25.0% with a 535 fold purification factor. The purified uricase migrated as a single protein band on reduced and non-reduced SDS-PAGE. The purity analysis by HPLC was above 97.5%. Native molecular masses was about 127 kDa, measured by Superdex 200 gel filtration chromatography. Reduced SDS-PAGE showed the molecular weight of 33 kDa. So, the uricase consists of four identical subunits. The enzyme exhibited maximum activity at pH 8.5 and 40℃. The Km of purified Uricase was 35.0μmol/L at pH 8.5 and 40℃.
