采用 ConA-FITC 直接荧光法和 ConA 间接免疫荧光法及 ConA-HRP 细胞化学法观察了实验肿瘤细胞(鼠乳腺癌 MA73和鼠肉瘤 S180)及临床标本人膀胱癌和结肠癌细胞的涂片标本。结果肿瘤细胞与 ConA结合,作为对照的正常细胞不与 ConA 结合。临床标本中癌细胞表面的 ConA 受体分布呈两种形式,一种均匀分布,另一种呈“斑块”状分布。实验中采用的肿瘤细胞直接涂片以 ConA—FITC 直接荧光检测法简便快速,60min 内可出结果,客观性强,协助病理诊断很有实用价值。

观察刺五加多糖(ASPS)对小鼠肉瘤S180细胞、人白血病K562细胞体外增殖的影响,发现ASPS对两种细胞的增殖均有强烈抑制作用,半数有效抑制浓度分别为0.38g·L-1和0.28g·L-1。为探讨其抗肿瘤机理,对S180细胞膜成分进行了分析,结果显示ASPS与细胞接触24h,引起细胞膜唾液酸含量升高,膜磷脂含量降低,膜磷脂脂肪酸组成发生明显改变(C20:0、和C14:0降低),与对照组相比,这些变化均有显著差异(P<0.05或P<0.01);但膜胆固醇含量、膜流动性(C/P比值、U/S比值)及膜中性脂脂肪酸组成不受影响。将ASPS与S180细胞膜在适当条件下一同温育,发现ASPS干扰膜的肌醇磷脂代谢,明显抑制磷脂酰肌醇转换。上述结果提示ASPS的抗癌机理与膜生化特性改变有关,其中对膜磷脂含量、脂肪酸组成和作为膜磷脂组分之一的肌酸磷脂代谢的影响是重要环节
