ObjectiveUsing a rat model of polycystic ovarian syndrome(PCOS), the role of transforming growth factor alpha(TGF-α) and its receptor were explored in the pathogenesis. MethodsProgesterone-synchronized rats were used as the model of PCOS by injecting human chorionic gonadotropin. TGF-α and its receptor were determined by immunohistochemical staining and ultrastructure of rats' ovaries were observed with transmission electron microscope. ResultsThe granulosa and theca cells of PCOS possessed ultrastructure features different from those found in normal ovaries. Immunostaining of TGF-α in the granulosa cells of PCOS was obviously weaker than that of normal controls (P< 0.01). Immunostaining of epidermal growth factor receptor(EGFR) in PCOS was obviously stronger than that of normal control(P< 0.01). ConclusionThe disruption of TGF-α and its receptor may be involved in the pathogenesis of PCOS.

目的利用多囊卵巢综合征 (polycysticovariansyndrome ,PCOS)大鼠模型 ,分析转化生长因子 α(transforminggrowthfactor alpha ,TGF α)及其受体在PCOS发病中的作用。方法采用皮下埋植左旋甲基炔诺酮联合腹腔注射人绒毛膜促性腺激素的方法建立PCOS大鼠动物模型 ,采用免疫组织化学方法检测TGF α及其受体在大鼠卵巢中的表达 ,并通过透射电镜观察PCOS大鼠卵巢超微结构的改变。结果PCOS大鼠卵巢颗粒细胞及泡膜细胞的超微结构明显不同于正常大鼠 ,PCOS组TGF α在颗粒细胞的染色比正常对照组明显减弱 (P <0 .0 1) ,而表皮生长因子受体 (epidermalgrowthfactorreceptor,EGFR)在卵巢各类中的染色均比正常对照组明显增强 (P <0 .0 1)。结论卵巢局部TGF α及其受体的调节失常是导致PCOS发生的重要因素之一
