At the concentration of 10 μg/ml, 7α-methyl-10β, 17β-diacetoxy-△~4-stren-3-one showed significant inhibiting action on progesterone biosynthesis in isolated pregnant rat ovary. Relative binding affinity (RBA) of 7a-methyl-lOft, 17β-diacetoxy-△~4-estren-3-one for rabbit uterine estrogen receptor was found to be 10.8 and that of 7β-methyl-10β, 17β-diacetoxy-△~4-estren-3-one to be 1.5. However, 7△-and 7β-methyl-10β, 17β-diacetoxy-△~4-estren-3-one were of poor affinity for progesterone binding sites. Result obtained from the uterine growth test in immature mice showed that the estrogen and antiestrogen activity of 7β-methyl-10β, 17β-diacetoxy-△~4-estren-3-one was weaker than that of 7α-methyl-10β, 17β-diacetoxy△~4-estren-3-one.

The anti-implantation action of norethisterone oxime (NETO) and that of norethisterone (NET) were compared in rat, hamster and rabbit. In addition, the progesterone concentrations in the ovarian vein of pseudo-pregnant rats receiving NETO or NET were measured by radioimmunoassay. The inhibition sction of NETO is also stronger than that of NET. The results suggest that oximation of ketone group of NET at C3 is able to enhance the anti-implantation action and the suppressive effect on the biosynthesis of progesterone in the rat ovary.

本文比较了炔诺酮肟与炔诺酮对大鼠,金地鼠及家兔3种动物的抗着床作用,用放射免疫法测定了两药引起大鼠卵巢静脉血中孕酮含量的变化。结果显示:随动物种属不同,炔诺酮肟的抗着床作用比母体炔诺酮强2~10倍。最小100%抗着床剂量的炔诺酮肟能抑制大鼠卵巢分泌孕酮,其作用比同等剂量的炔诺酮明显增强。提示炔诺酮A环三位酮基肟化能提高炔诺酮的抗着床作用,并增强其对卵巢孕酮生物合成的抑制。

Methods The change of ultrastructure of ovary and testis after treating orally with 4.6mg/kg/d of cyproheptadin was observed under the transmmission electronic microscope. Results The mitochondria of follicular cells and corpus luteum of female rats were swollen, cristae fractured, and myelonic and vacuolar in the ovary in experimental group. And degranulation also appeared in rough endoplasmic reticulum in follicular cells, while no obvious change was found in testis. Conclusion Cyproheptadine caused the obvious changes of the ultrastructure of the follicular cells and corpus luteum of female rats.

目的 研究赛庚啶 (cyproheptadine,CYP)对大鼠卵巢和睾丸超微结构的影响。方法 用透射电子显微镜观察经 4 6mg/kg/d赛庚啶灌胃后对卵巢和睾丸组织超微结构的改变。 结果 试验组大鼠卵巢中卵泡细胞和黄体细胞线粒体肿胀、嵴断裂 ,出现髓样变 ,甚至呈空泡样 ,卵泡细胞粗面内质网脱颗粒。睾丸组织未见明显改变。结论 赛庚啶对雌性大鼠卵泡细胞和黄体细胞超微结构产生明显影响。
