Objective To study the expression of bcl- 2? p16 proteins and their relationship in childhood acute leukemia, and to discuss the role of apoptosis and proliferation in the pathogenesis of acute leukemia. Methods The expression of bcl- 2? p16 proteins in bone marrow biopsy issues were measured by immunohistochemistry staining in 29 cases of newly diagnosed childhood acute leukemia selected as test group and in 19 cases without leukemia served as control group. Results The expression of bcl- 2 protein in test group was significantly higher than that in control group. The expression of p16 protein in test group was significantly lower than that in control group. There was a negative correlation between bcl- 2 and p16 expression in childhood acute leukemia. Conclusion Both apoptosis inhibited by bcl- 2 protein high expression and proliferation enhanced by p16 protein deletion might play an important role in the pathogenesis and progression of acute leukemia, and the role of apoptosis inhibition might be heavier than proliferation enhancement. 目的研究初诊儿童急性白血病bcl-2、p16蛋白的表达及两者关系,探讨细胞凋亡和细胞增殖在急性白血病发生中的作用。方法采用免疫组化技术检测bcl-2和p16蛋白在29例初诊急性白血病患儿骨髓活检组织中的表达,并与19例非白血病患儿的表达作对照。结果实验组bcl-2蛋白表达显著高于对照组;实验组p16蛋白表达显著低于对照组;在儿童初诊急性白血病中p16与bcl-2表达呈负相关。结论bcl-2过度表达引起的细胞凋亡抑制增强和p16失表达引起的细胞增殖抑制减弱在急性白血病发生发展中发挥重要作用;细胞凋亡障碍和细胞增殖失控在多数白血病发病中可能存在某种协同作用,细胞凋亡抑制在部分白血病的发病中可能比细胞增殖失控起的作用更重要。
