The effect of AMG- 1 [6( 5 - hydroxy- 2- methylpyridylamino ) - 9 ribofranosylpurine] on mouse brain energy metabolism in complete brain ischemia induced by decapitation and on neurological deficit and histopathological changes after occlusion of the middle cerebral artery (MCAO) in rats were studied. The results indicate that AMG-1 has an effect of improving the status of energy metabolism in complete brain ischemia in mice. The effect of AMG- 1 [6( 5 - hydroxy- 2- methylpyridylamino ) - 9 ribofranosylpurine] on mouse brain energy metabolism in complete brain ischemia induced by decapitation and on neurological deficit and histopathological changes after occlusion of the middle cerebral artery (MCAO) in rats were studied. The results indicate that AMG-1 has an effect of improving the status of energy metabolism in complete brain ischemia in mice. Lactate concentration was greatly reduced and the ATP and phosphocreatine levels were significantly elevated. Treatment with AMG-1 or nimodipine was performed before and after MCAO. The score of neurological deficit was significantly decreased as compared with the vehicle treated group. The extent of ischemic neuronal injury was determined by histopathological examination. AMG- 1 and nimodipine seemed to attenuate the MCAO - induced neuronal damage. In as much as AMG- 1 can improve the status of brain energy metabolism, neurological deficit and neuronal damage after ischemic insult, AMG- 1 may have a beneficial effect for the treatment of cerebral ischemic damage.

Beta-endorphin immunoreactive substance ( ir beta-endorphin ) content were measured in cerebrospinal fluid at pre-ischemia and after ischemia and reperfusion stages. In control group, the content of ir beta-endorphin significantly increased (1.01±0.52 mg/L, P< 0.05). In phencyclidine 20μg/kg icv treated group, the content of ir beta-endorphin increased slightly ( 0.36 ± 0.32 mg/L) during ischemia and reperfusion, however in phencyclidine 80μg/kg icv treated group, the content or ir beta-endorphin decreased (0.19±0.44 mg/L), which was significant difference from control group ( P<0.05 ). These results indicate that phencyclidine may antagonize the centrally released beta-endorphin induced by acute cerebral ischemia and show benefical effects in cerebral ischemic damage.

结扎兔双侧椎动脉和颈总动脉,导致急性脑缺血。缺血30min后重新开放双侧颈总动脉,使再灌注。脑缺血30min再灌注2h后,脑脊液中β—内啡肽免疫活性物质含量比缺血前增加1.01±0.52mg/L。icv苯环利定20μg/kg组,脑脊液中β—内啡肽免疫活性物质含量比缺血前增加0.36±0.32mg/L。icv苯环利定80μg/kg组,脑脊液中β—内啡肽免疫活性物质含量比缺血前下降0.19±0.44mg/L。提示苯环利定能抑制脑缺血后脑内β—内啡肽释放,减轻脑缺血性神经损伤。

Clavirolide C is a unsaturated lactone diterpenoid isolated from Clavularia viridis Quoy and Gaimard. Pharmacological experiments showed that clavirolide C ip 1.2mg/kg decreased malondialdehyde (MDA) and elevated activity of superoxide dismutase (SOD) in cerebral ischemic rats.9 and 18 μmol.L~(-1) clavirolide C inhibited ADP or Ca~(2+) induced rabbit platelet aggregation rates.These resuIts suggest that clavirolide C play an protective role in cerebral ischemic damage of rats.

绿柱虫γ-内酯二萜(简称C)显著降低脑缺血组织脂质过氧化产物丙二醛的含量,提高超氧化物歧化酶活性,从而发挥其对脑缺血再灌注损伤之脑组织的保护作用。对脑组织的保护作用还与它能显著抑制血小板聚集有关。结果表明,C对缺血脑组织的保护作用可能在于它能拮抗Ca~(2+),从而减轻再灌注时Ca~(2+)所造成的损伤。
