Peripheral lymphocytes were purified from 114 cancer patients, 48 noncancer patients and 30 normal adults. Tumor antigen, incubated with lymphocytes from a corresponding cancer patient, produced a significant increase in the ability of the lymphocytes to form active rosette-forming cells (A-RFG) when compared with lymphocytes...

The active rosette test was adapted as in vitro assay and researched for relation withhuman tumor antigens——gastric cancerous, lung cancerous and hepatic cancerous. Peripheral lymphocytes were purified from 114 cancer patients, 48 noncancer patients and 30 normal adults. Tumor antigen, incubated with lymphocytes from a corresponding cancer patient, produced a significant increase in the ability of the lymphocytes to form active rosette-forming cells (A-RFG) when compared with lymphocytes cultured without antigen (p∠0.001). This increasing effect on A-RFC still remained after refrigeration. Tumor antigen incubated with lymphocytes from normal donors produced no increase in their A-RFG. It should be noted that this assay appears in a certain manner to be specific for the antigen to which the individual is corresponding cancer patient, for example, gastric cancerous antigen activated A-RFG of a gastric cancerous patient, but produced no increase in breast patient. This assay provides a new in vitro method for investigating mechanism for cell-mediated immunity and also a new specific diagnostic method for cancer.

This paper reports the study of E and EAC rosette formation test with the use of spleen cells from inbred 615 mice. Twenty-four normal 615 mice of 20-22 gm in body weight were tested. The numbers of E and EAC rosette forming cells in their spleen cell suspensions ranged between 21.3± 7.1% and 38.5±9.4% respectively. There was no significant difference between male and female mice. Several factors which might possibly influence the experimental results were discussed.

A study was made for investigating the significance of esterase staining in differentiation of T and non-T lymphocytes. The lymphocytes isolated from the peripheral blood of 26 normal cases were studied with esterase staining, and corresponding rosette tests were made for ascertaining the relationship of these two methods.The results obtained were as follows: 1. 99.3±2.1%(X±SD) of active ERFC (E rosette forming cell) and 94.4±7.2% of total ERFC were found to be ANAE(+). 2. 82.4±5.6% of ANAE(+) lymphocytes exhibited the rosette forming ability. 3. The percentage of total ERFC was 60.9±8.5%, while that of ANAE(+) lymphocytes was 56.5±10.5% (12 normal cases).There was no significant difference (P>0.05). It is apparent that the ANAE activity is always coincident with E rosette forming ability, and both of them will serve as indicators of T lymphocytes. Thus under well-controlled experimental conditions esterase staining will find wide applications in the assay of cellular immunity due to its simplicity and feasibility.

为探讨酯酶染色技术在鉴别T与非T淋巴细胞上的意义,从正常人(26例)外周血分离淋巴细胞,作酯酶染色,同时与E花结试验作对比,以研究两种技术的相关性。结果如下: 1、99.3±2.1%(均值±标准差)的活性ERFC(E花结形成细胞),94.4±7.2%的总ERFC均呈ANAE(+)。 2、ANAE(+)淋巴细胞中,82.4±5.6%具有形成E花结的能力。 3、总ERFC百分率为60.9±8.5%:ANAE(+)淋巴细胞(12例)百分率为56.5±10.5%,二者的差别不显著(P>0.05)。由上可知,ANAE活性和E花结形式成能为常平行存在于T淋巴细胞,二者均可作为T淋巴细胞的标志,酯酶染色由于其方法简便,如能严格控制实验条件,在细胞免疫测定中将获得广泛应用。
