A high-performance liquid chromatography (HPLC) method has been proposed for the determination of imidacloprid residues in fortified potato (Solanum tuberosum L.) and onion (Allium cepa) and in treated potato and onion samples.
      
Sample preparation consisted of dichloromethane extraction of imidacloprid from the plant material, followed by purification of the obtained onion extract on an LC-Florisil disposable cartridge.
      
The HPLC-diode-array-detection (DAD) method has been developed on reversed phase for separation of imidacloprid with isocratic elution with a mixture of 0.01 M phosphate buffer (pH 3.15) and acetonitrile (75 : 25, v/v).

In this paper, a new method to assess the environmental safety of pesticide has been proposed in terms of sperm deformity of the earthworm (Eisenia foetida). The results showed that the main sperm morphological deformity appeared to be shape changes of the sperm heads. Significant induction of sperm deformity (P<0.01) and dose effect relationship displayed when the earthworms were exposed to imidacloprid at the concentrations of more than 0.5mg/kg dry soil. However, The sperm deformity frequency of RH-5849-exposed groups did not show significant difference (P<0.05) from the negative control until the dose reached 100 mg/kg dry soil. This study suggests that the detection of sperm abnormality in earthworms could provide an early warning system of polluted environment.

本文提出了一种检测及评价土壤生态系统中遗传毒性的新方法。赤子爱胜蚓(Eiseniafoetida)经人工土壤法染毒,暴露于不同剂量的两种新型杀虫剂———呲虫啉和抑食肼中,观察它们对蚯蚓精子形态的影响。实验结果表明,呲虫啉在浓度大于0.5mg/kg干土时对蚯蚓精子畸变率有明显影响(P<0.01),并呈现剂量效应关系;而抑食肼在浓度高达100mg/kg干土时,与对照组相比才有显著差异(P<0.05)。

Objective To study the effect of new-typed pesticide imidacloprid on the content of NO(nitric oxide) in nutrient solution and the activity of NOS (NO synthase) in primary-cultured rat, hippocampus neurons.Methods To poison mature primary-cultured rat's hippocampus neurons with imidacloprid of different concentration; then to observe the morphological change under phase difference microscope to determine cell mortality,the content of NO in nutrient solution and the activity of NOS respectively at time of 12h,24h and 48h after poisoning.Results Compared with DMSO control group, there are following changes for each group apart from groups of zero and 10 -6 mol/L imidacloprid with increase of poisoning concentration (10 -5 -10 -3 mol/L) and prolonging of poisoning time.Hippocampus neurons become swell,the intracellular granules increase,the axon and dendrite break or even loss,the intercellular network also becomes sparser.The cell mortality and the content of NO increase after poisoning for 24h ( P <0.05),and they increase much more at time of 48h ( P <0.01).The activity of intracellular NOS of 10 -3 mol/L imidacloprid group increases at time of 12h( P <0.01),and it becomes higher at 24h ( P <0.01),that of 10 -5 -10 -4 mol/L imidacloprid groups also increases at time of 24h( P <0.01).And there is no obvious change for the activity of NOS of every group between time of 24h and 48h.Conclusion Imidacloprid with concentration range from 10 -5 mol/L to 10 -3 mol/L can result in increase of the content of NO in nutrient solution and the activity of NOS in primary-cultured rat's hippocampus neurons,which may be related to its neurotoxic effect.

In the comet assay, starting from the lowest dose the two pesticides caused DNA damages in the human peripheral lymphocytes as compared with the control groups, and the differences were significant (P<0.01), and there were dose dependent relationship (r=0.995, r=0.965).Conclusion: Both imidacloprid and RH 5849 damaged the DNA of human peripheral lymphocytes. Comparatively. Imidacloprid was more toxic than RH 5849.
