A full length cDNA of the coding region of hTPO,whose sequence is identical to those previously reported,has been amplified by means of RT PCR with human fetal liver mRNA as the templates and then sequenced.Further,this insert was subcloned into a mammalian expression vector pcDNA3 (pcDNA3 TPO),transformed into CHO cells, and screened by G418 and then tested by expression level of hTPO.A group of stably secreting engineered strains of hTPO has been obtained.

The encapsulation of living cells would be useful as transplanted tissue without the danger of immune rejection. Our studies on microencapsulation and transplantation of chromaffin cells clearly show that this is the most promising approach to the problem of Parkinson's disease and cancer pain. The results of these studies will make the clinical application of our concept of chromaffin cells transplantation...

Since 1980 a number of potential medical applications of microencapsulated cells have been developed. The encapsulation of living cells would be useful as transplanted tissue without the danger of immune rejection. Our studies on microencapsulation and transplantation of chromaffin cells clearly show that this is the most promising approach to the problem of Parkinson's disease and cancer pain. The results of these studies will make the clinical application of our concept of chromaffin cells transplantation possible in the near future. It is clear that transplantation of mammalian cells or genetically engineered cells encapsulated in protective, biocompatible, and semipermeable membranes has great clinical potential for a wide range of diseases requiring enzyme or endocrine replacement therapy.

Objective To observe the role of genetically engineered myoblast in the treatment of rat Parkinson disease models Methods Genetically modified myoblasts to produce brain derived neurotrophic factors (BDNF) were established and grafted into the corpora striata of Parkinson disease modelsin rats. The behavior and pathological changes were observed and the Dopamine contents in the corbora striata were measured with high performance liquid chromatography (HPLC). Results The intracranial graft of genetically engineered myoblasts can promote the survival of TH positive neurons in the nigra of Parkinson's rat and the content of Dopamine in the striatum were increased significantly. The rotation behavior improved 50% after the transplantation. Conclusion It is suggested that BDNF secreting myoblasts can promote the survival of the neuron and provide a new and effective method for the treatment of Parkinson disease.

目的观察脑源性神经营养因子(BDNF)基因工程成肌细胞脑内纹状体移植对帕金森病大鼠的治疗作用。方法建立逆转录病毒介导的BDNF表达质粒并转染成肌细胞,筛选阳性细胞进行脑内移植。结果基因工程成肌细胞脑内移植可明显提高帕金森病大鼠黑质酪氨酸羟化酶(TH)阳性神经元的存活率,使纹状体多巴胺含量明显增加,动物的旋转行为改善约50%,并持续2个月之久。结论脑源性神经营养因子基因工程成肌细胞脑内纹状体移植可明显改善动物的旋转行为并可促进黑质神经原的存活,为帕金森病的治疗提供了一种新的有效的治疗方法
