Successful transplantation of the fetal brain tissue into a resection cavity on the motor cortex area of young, adult and aging host brain is described. These studies explored the survival, growth and viability of the grafts from El2, El5 and El8 rats fetal frontal cortex (door and host wistar rat). Animals were prepared for light microscope (Nissl Golei)2, 4 and 6 months later .Implants by both fetal tissues had a 80% survival rate. In motor cortex of the host brain, the younger the fetal implant, the higher the success of the implant, but the most successful implants were the E15 (El5>El2>El8) . For the speed of the growth the El2 is the fastest (El2>El5>El8).The implants not only increased in mass (up to 6—7 fold in most of cases at 4 months) but differentiated and matured neurons were observed one to two months postimplantation.Implants were easily identified in all cases on the basis of cellular organization and the presence of a neuron sparse Zone present at the borders, just like the molecular layer of the normal cortex.There was no apparent difference in the size or organization of implants examined 4 moths after implantation as compared with those examined after longer survival times. Golgi staining demonstrated that both pyramidal and nonpyramidal cells were in the grafts. No preferential positioning or lamination of these different cells were seen with respect to each other, although some pyramidal cells had apical dendrites oriented toward "molecular zones" . These had the most recognizable pyramidal morphology, although pyramidal cells through the graft had many typical aspects of pyramidal cells such as large "apical" dendrites and spines.Nonpyramidal cells were of two types spiny and aspiny.

Objective To compare the effects of combined fetal spinal cord allograft and vascularized or free peripheral nerve autograft to repair spinal cord injury in adult rats. Methods A total of 30 adult, female Wistar rats were used in this study, and a 5 mm cord defect of the left lateral column was made at T 1~3 vertebral level. The defect was grafted randomly using embryonic day 14 fetal spinal cord and vascularized (Group V+F), or free peripheral nerve(Group P+F) respectively. Eight weeks after surgery, the survival, differentiation of the grafts, and the ability of repairing host spinal cord were assessed by histological, histomorphometric analysis and somatosensory evoked potential (SEP) studies. Results In group V+F, the fetal implant was intimately fused with the host CNS parenchyma. The volume, nerve fiber and neurons density of the fetal implant were significantly higher than Group P+F ( P <0.05). Grafted fetal spinal cord exhibited a high level of maturity, SEP assessments showed that the latency of P 1, N 1 wave was significantly decreased in this group ( P <0.01). Conclusions Group V+F has an advantage over group P+F in histological and SEP, and this combination could promote the development of fetal implant and support the regeneration of central injured axons to certain degrees.

目的 比较胚胎脊髓与带蒂或游离周围神经联合移植修复脊髓损伤的能力。 方法  30只成年大鼠损伤胸髓后行胚胎脊髓加游离正中神经 (P +F组 )或加带蒂正中神经 (V +F组 )移植。术后 8周行体感诱发电位 (SEP)和光、电镜检查。 结果 V +F组胚胎脊髓与受体融合较好 ,体积增长 378.4%、神经微丝蛋白密度 (7.46± 0 .77)万个 /mm2 、神经元密度 (1.36± 0 .34)万个 /mm2 ,均高于P +F组 (P <0 .0 5 ) ,细胞分化程度高 ,SEP的P1、N1波潜伏期显著缩短 (P <0 .0 5 )。 结论 胚胎脊髓与带蒂神经联合移植 ,能促进胚胎脊髓成熟分化和损伤神经元再生 ,效果优于与游离周围神经联合移植

The defects were grafted respectively 14 days em bryonic fetal spinal cord (Group FSC,15 animals ) or vascularized peripheral nerve and fetal spinal cord (Group V+F,15animals),and another 6 animals were kept as the control group.8 weeks after surgery,the survival,differentiation of the grafts,and the ability of repairing host spinal cord were checked.Results:In the cograft group,the fetal implant was intimately fused with the host CNS parenchyma,few glial scar partition were presence.The volume,nerve fibers and neurons density of the fetal implant were significantly higher than Group FSC(P<0.01).The majority of neurons in this group were well differentiated,including some clusters of large neurons resembling ventral horn cells of the intact spinal cord,well developed synapses were scattered throughout the neuropil.SEP showed that the latency of P 1 and N 1 waves was significantly decreased in this group (P<0.01) .Conclusion:The combination method could prompt the development of fetal implant and the regeneration of central injured axons to certain degrees.

目的 :探讨神经组织联合移植对成鼠急性脊髓损伤的修复能力。方法 :成年雌性Wistar鼠 36只随机分为3组 ,损伤T1~ 3脊髓左后柱 ,移植孕 14d胚胎脊髓 (FSC组 15只 )或带血管正中神经加胚胎脊髓 (V +F组 15只 ) ,另 6只做对照组。术后 8周行体感诱发电位、光、电镜和免疫组化检查。结果 :V +F组胚胎组织体积增长速度、神经纤维和神经元数目显著高于FSC组 (P <0 .0 1) ,细胞大多分化较好 ,有少数类似运动神经元 ,突触较成熟。界面区无明显胶质增生 ,SEP示P1、N1波潜伏期显著缩短 (P <0 .0 1)。结论 :带血管周围神经与胚胎脊髓联合移植优于单一移植物 ,对胚胎脊髓的生长发育、对损伤神经元的再生能力均有促进作用
