Radioimmunodetection in nude mice bearing human colon cancer cell line LS174T xenograft was performed in this article with hand-held gamma detector to provide reference for clinical RIGS McAb CL-3 was prepared by fusing of Sp2/0 myeloma cells with spleen cells of BALB/c mice immunized with human colon cancer cell line LS174T.CL-3 F(ab')2 was prepared by pepsin digestion of CL-3. The antibodies and fragments were iodinated with 125I using iodogen method. The two groups of mice were detected with HHGD at 24,48,72,96,120, 144,168th hour after injection of radiolabelled antibodies and fragments. The T/N in 125I-CL-3 group yaned between 1.22~3.79, with the highest level at 168th hour; while the T/N in 125I-CL-3 F(ab')2 group was 2.0~4.5,the highest being at 72~144th hour. The results indicate that after intraperitoneal injection of 125I-CL-3 and 125I-CL-3 F(ab')2,the radiolabelled antibidies anb fragments can specifically concentrate in tumor xenografts and can be detected by HHGD,and that F(ab')2 fragmens are better in localization than intact antibodies.

本文以手持式γ探测仪对荷人结肠癌细胞系Ls174T裸鼠进行体表放射免疫探测,为临床应用放射免疫引导外科手术(RIGS)提供了依据。以人结肠癌细胞系Ls174T免疫BALB/c小鼠制备McAbCL-3,胃蛋白酶消化法制备CL-3F(ah′)2片段,Iodosen法以125I标记CL-3及其F(ab′)2。两组荷瘤裸鼠注射标记物后分别于24、48、72、96、120、144和168h以手持式γ探测仪(HHGD-I型)进行体表探测,测定T/N比值。125ICL-3组T/N比值在1.22~3.79之间,第168小时最高;125ICL-3F(ab′)2组T/N比值在2.0~4.5之间,第72~144小时最高。结果表明,荷瘤裸鼠腹注腔射125ICL-3及125ICL-3F(ab′)2后标记物能特异性地浓集于肿瘤组织,以手持式γ探测仪进行体表探测T/N比值有明显意义,能明确定位肿瘤。F(ab′)2用于RIGS裸鼠实验效果优于完整抗体。

Radioimmunoguided surgery (RIGS) is anew branch of surgery that rose in 1980s. With the devel-opment of biomedicine, tumor monoclonal antibody is ap-plied to clinical surgery from experimental study gradually.Being infused into a body, the monoclonal antibody labeledby radioactive isotope will combine with the antigen, which exist on the surface of tumor cell. The tissues gathered withmonoclonal antibody can be detected by portable gamma -dctector during the operation hecause of the radioactive raysmay be emitted from these tissues where tumor probablyexists. According to the study of E. W Martin from theMedical College of the State University of Ohio, USA, thelatent tumor, which is diffcult to find only by visual senseand hands, could he detected with this new technology.About thirty percent of the operations could be improved,due to portable gamma - detector and it's particularly ef-fective to the patients with recurrent tumor. Our study was to investigate the effectiveness of radioimmunoguided surgery (RIGS) in lung carcinoma xenografts by using monoclonal antibody C50 labeled with 99m Tc. 40 mice bearing lung adenocarciomona models were divided into two groups according to presence or absence of lung metastases, and were injected with a dose of 200uCi/40ug of 99m Tc C50 into the tail veins.The external radioimmunoimagings were performed after injection. Our study was to investigate the effectiveness of radioimmunoguided surgery (RIGS) in lung carcinoma xenografts by using monoclonal antibody C50 labeled with 99m Tc. 40 mice bearing lung adenocarciomona models were divided into two groups according to presence or absence of lung metastases, and were injected with a dose of 200uCi/40ug of 99m Tc C50 into the tail veins.The external radioimmunoimagings were performed after injection. AⅡ viscera and tumor were detected in vivo by means of a hand held gamma detecting probe (GDP), and the radioactive distribution was analyzed by employing well gamma detector. The result showed that the tumor images of mice were clear, and the images were best at six hours after injection.The values of %ID/g of tumor and kidney were higher than those of other organs.Ratios of tumor/normal tissue(T/NT) detected by GDP ranged from 2.23 to 9.73.T/NT ratios of lung metastases were 2.09±0.31. The study suggests that C50 labeled with 99m Tc has a good specificity and affinity to primary lung neoplasms as well as lung metastases, thus the neoplastic tissues could be identified accurately from normal tissues and the micrometastases could be detected by using GDP.

本研究以核素99mTc标记抗癌胚抗原 (CEA)单抗C5 0对肺癌模型的放射免疫导向手术 (RIGS)进行初步的探讨。根据有无肺转移 ,40只荷肺腺癌小鼠被随机配对分成两组 ,并经尾静脉注入 2 0 0 μCi/ 40 μg99mTc C5 0。注药后进行体外放免显像、手持型γ探测仪 (GDP)体内探测及井型探测仪分析。结果显示注药后 6h ,肿瘤显像最清晰。井型探测仪探测的肾脏与肿瘤的 %ID/ g高于其它脏器。GDP探测的肿瘤组织与正常组织的放射性比值 (T/NT)范围为 2 .2 39.73。肺转移癌的T/NT值为 2 .0 9± 0 .31。实验表明 ,99mTc C5 0对肺癌原发病灶及转移灶有较高的特异性和亲和力 ,利用GDP可准确区分肿瘤及正常组织并探测出微小转移病灶。
