Objective This study detected the expression of Skp2 in epithelial ovarian tumors and analyzed the relationship between its expression levels and clinicopathological parameters in ovarian adenocarcinomas. The purpose is to explore its role in the tumorigenesis and development of epithelial ovarian tumors and to identify its significance in evaluating the prognosis of ovarian adenocarcinomas.Methods Expression of Skp2 was examined by immunohistochemistry in 81 epithelial ovarian tumors [13 adenomas,12 borderline adenomas,and 56 adenocarcinomas]. Results of immunohistochemistry were correlated with clinicopathological variables.Results Expression of Skp2 was detected in neither ovarian adenomas nor borderline adenomas.In contrast, it was detected in 46.43% (26/56) of adenocarcinomas. Positive expression of Skp2 was detected significantly more often in adenocarcinomas than that of borderline adenomas (P< 0.0001),which was significantly more often than that of adenomas (P< 0.0001) in turn. Counting the percentage of positive cells of 26 adenocarcinomas with Skp2 positive expression indicated that the expression level of Skp2 was significantly higher in advanced-grade specimens compared with early-grade ones(P<0.01).In addition,the level of Skp2 expression also significantly associated with clinical stage(P<0.01)and positive lymph node metastasis (P< 0.05)of ovarian adenocarcinomas.Conclusion Skp2 expression may play an important role in the development and progression of ovarian adenocarcinomas, and Skp2 expression can be used as a assistant marker of estimating the prognosis of ovarian adenocarcinomas patients.The higher expression levels indicates poorer prognosis.

目的 检测S期激酶相关蛋白2(S phase kinase associated protein2, Skp2)在卵巢上皮性肿瘤中的表达,探讨其表达与上皮性卵巢癌临床病理参数之间的关系,以及了解其表达在卵巢上皮性肿瘤发生、发展及上皮性卵巢癌预后估计中的意义。方法 收集我院81例卵巢上皮性肿瘤的外科手术石蜡标本,用免疫组化的方法检测Skp2的表达水平。结果 Skp2 在卵巢上皮性良性肿瘤及交界性肿瘤中表达均为阴性,而在上皮性卵巢癌中表达率为46.43%(26/56),半定量分析表明Skp2 在不同年龄及组织类型的卵巢癌中表达无显著性差异(P>0.05),而在中低度分化组、Ⅲ~Ⅳ期组及有淋巴结转移组分别明显高于高分化组(P<0.01)、Ⅰ~Ⅱ期组(P<0.01)及无淋巴结转移组(P<0.01)。结论 Skp2 特异性表达于上皮性卵巢癌,其表达水平越高提示病人预后越差。

Objective To investigate the expression of cyclin D1 and Ki-67 in epithelial ovarian neoplasm and its clinical significance. Methods Immunohistochemistry method was used to examine the protein expression of cyclin D1 and Ki-67 in 30 benign ovarian adenomas, 20 borderline tumors and 80 ovarian carcinomas, so as to analyze their correlations and asociation with patient’s clinical features. Results In this study, the positive rate of expression of cyclin D1 in benign ovarian adenomas, borderline tumors and ovarian carcinomas was 3%, 25% and 26%, respectively. In Ki-67 staining, 15% borderline tumors were detected overexpression of Ki-67; 39% ovarian carcinomas were observed Ki-67 overexpression. In addition, a significant association of Ki-67 expression with tumors Silverberg’s grade was evaluated (P<0.05), where 47% of carcinomas in G2/G3 were observed overexpression of Ki-67, the rate was obviously higher than that in G1 grade(17%). Conclusions Cyclin D1 and Ki-67 protein might be taken as a molecular marker to predict the benign or malignant potential of epithelial ovarian neoplasm; the overexpression of Ki-67 could be thought as an index to judge the cellular malignancy of ovarian carcinoma.

目的探讨卵巢上皮性肿瘤中cyclinD1和Ki-67蛋白表达及其临床意义。方法运用免疫组化方法,检测cyclinD1和Ki-67蛋白在30例卵巢腺癌、20例卵巢交界性肿瘤和80例卵巢癌中的表达情况,结合肿瘤的临床资料,分析其临床意义。结果cyclinD1蛋白在卵巢良性腺瘤、交界性肿瘤和卵巢中的阳性表达率分别为3%、25%、26%。Ki-67蛋白在交界性卵巢肿瘤的过度表达率为15%,在卵巢癌中为39%,而且与肿瘤Silverberg氏分级有显著的相关性(P<0·05),其中47%的G2/G3级的卵巢癌出现Ki-67蛋白过度表达,其过表达率明高于G1级的肿瘤(17%)。结论cyclinD1和Ki-67蛋白可以作为临床判断卵巢上皮性肿瘤良恶性有参考价值的分子标记;卵巢癌中Ki-67蛋白过度表达可成为评估肿瘤细胞恶性程度的指标之一。

Background and purpose:There are many oncogenes,such as FGF3 and cyclinD1,located at human chromosome 11q13.At present,however,the action of FGF3 abnormalities in epithelial ovarian neoplasm is unclear.The purpose of this study was to investigate the amplification and expression of FGF3 in epithelial ovarian neoplasm and its clinico-pathological significance.Methods:Immunohistochemistry(IHC) and Fluorescence In Situ Hybridization(FISH) methods were used to examine the protein expression and amplification of FGF3 in 30 benign ovarian adenomas,20 borderline tumors and 80 ovarian carcinomas,so as to analyze their correlations and association with patient's clinico-pathological features.Results:In this study,16% ovarian carcinomas were observed to have positive expression of FGF3,while none of the ovarian adenomas and borderline tumors were detected to be FGF3 positive.In addition,in ovarian carcinomas of this study,a significant association of FGF3 expression with patients FIGO stage was evaluated(P<0.05),where 23% of carcinomas in FIGO stage Ⅲ/Ⅳ were observed to be FGF3 positive,the rate was obviously higher than that in FIGO stage I/II(4%).In FISH study,only 1% of ovarian carcinomas were detected to have amplification of FGF3.None of the borderline and benign ovarian tumors were shown to have amplification of the gene.Conclusions:The positive expression of FGF3 might be taken as a molecular marker to predict the malignancy of epithelial ovarian neoplasm;the up-regulated expression of FGF3 might be involved in the invasive/metastatic process of ovarian carcinomas.The amplification of FGF3 gene is not the predominant mechanism of increased expression of its encoded protein in epithelial ovarian neoplasm.
