The regeneration of corneal nerves were observed after experimental penetrating keratoplasty in 40 rabbits by the cholinesterase histochemistry. It has been demonstrated that the first signs of regenerating axons were seen 1 week postoperatively in the severed end of nerves. After 4 weeks several single regenerating axons extended into the graft through he scar between host and graft, while a teural proliferating reaction were apparnantly observed in the host cornea. The new neurites and the regenarating fibers all grew in the direction of the graft so that the neural density significantly incre ased in the host and near the scar. 6-10 weeks after the operation the number of regenerating fibers were increased in the peripheral graft, but only a few fine regenerating axons were seen in central part in spite of 3 months after the operation.

The corneal nerve regeneration and corneal sensation recovery following corneal surgery have been a difficult problem. So far, however, the origins of corneal afferent nerves related to nerve regeneration have not been confirmed. For this reason, we studied the origins of SP-LI fibres and CGRP-LI fibres of central area of cornea of rat by using the combined horseradish peroxidase(HRP) retrograde tracing and immunohistochemistry method. Solution of 30% HRP was injected into the corneal epithelium of central area of rat cornea under stereomicroscope. After a survival period of 2 days, the rats were perfused transcardially with 4% paraformaldehyde. The trigeminal ganglion (TG). superior cervical ganglion (SCG) and vagus ganglion (VG) were removed and frozen-sectioned serially at 40um. The sections were processed according to TMB method to show HRP labelled cells, then the sections were treated with immunohistochemistry method and examined by bright-field microscopy. The resuhs were as follows: double labelled neurons were found in the medial area of the ipsilateral TG (ophthalmic region) and concentrated at the dorsal part of the area. The CGRP-LI neurons were distributed in the peripheral part of dorsal-medial area of TG. The SP-LI neurons were scattered in the deeper pan of dorsal-medial area of TG. The shapes of SP-LI and CGRP-LI neurons are round or elliptical. There were no double labelled neurons in the SCG and VG.This experiment not only confirmed the hypothesis that the corneal SP-LI fibres originated from TG neurons but alro proved that the double labelled neuron was not found in the SCG and VG which suggested that the SP-LI fibres and CGRP-LI fibres were not come from neuorns of SCG and VG.

向角膜中央区注射HRP后,用抗SP和抗CGRP抗体对三叉神经节、颈上神经节和迷走神经节进行免疫组织化学处理。结果证明:双重阳性细胞出现于三叉神经节的内侧区且集中分布于背内侧部.CGRP样双重阳性细胞主要分布在节的背内侧部的周边区;SP样双重阳性细胞散在于节的背内侧部的深部。两者均为圆形或椭圆形。在颈上神经节和迷走神经节内未见双重阳性细胞。
