Fifty human fetal pancreases of 4-6 months gestation obtained from legal abortions were microenoapsulated with alginate, poly-lysine after culture for 4-7 days. The mierocapsules were studied morphologically and functionally.Immunooytoohemical staining for insulin indicated β-cells were morphologically intact at 48 days. No exicrube function was detected as evidenced by the low amylase levels.It is concluded that mioroenoapsulated human fetal pancreatic tissue retains viability for more than 25 days and may be expectantly useful as transplants in the treatment of diabetes mellitus.

作者对50例主要取自水囊引产的4至6月龄人胎的胰腺,在机械分散、体外培养4至7d后,用海藻酸钠、多聚赖氨酸等高分子生物材料进行微囊化,并在体外培养条件下采用免疫酶标组织化学染色(PAP法)、放射免疫测定法、微量氧化酶法,研究微囊化人胎胰组织的存活、分化、增殖情形及激素和外分泌酶分泌功能。结果表明:微囊化人胎胰组织体外培养48d,β细胞分化增殖良好。培养条件下25d仍保留有良好的胰岛素、C肽分泌功能,与未微囊对照组织无明显差别。未见明显的淀粉酶分泌功能。实验提示微囊化人胎胰组织可望成为一种有前途的移植物,供临床治疗胰岛素依赖性糖尿病。

Islet transplantation was performed in fourteen patients sufferingfrom insulin dependent diabetic mellitus for 1~11 Years. Before tranplantation the patients were treated with insulin 22~60 units/day and ketoacidosis appeared recurrently in 4 cases. Human fetal pancreatic tissue was transplanted in the omental bursa and the patients were followed up for 6~20 months. 12 cases (85.71%) were found effective and the daily dosage of total insulin was decreased by more than 305%. 14例Ⅰ型糖尿病,病程最短1年,最长11年。胰岛素用量每日22~60单位,其中4例反复发作酮症酸中毒。14例患者均系病情控制后,应用人胎胰岛行大网膜囊腔内移植,并随访6~20个月,发现12例有效,胰岛素每日用量减少30%以上。10例在移植10~14天时,有明显低血糖反应。研究表明,冷冻复苏胰岛细胞和未经冷冻胰岛细胞功能相似,光镜下两者组织细胞均生长良好。
