By immunizing Balb/c mice with immunoglobulin IgG which was isolated from the D_3 monoclonal antibody specific to the retinal ganglion neuronotrophic factor (RGNTF-McAb) with a Phast System gel electrophoretic method, we have developed anti-idiotypic antibodies against the RGNTF-McAb. A MTT colorimetric microassay designed to measure the effect of conditioned media from hybridomas containing antibodies on the growth activities of retinoblastoma (Rb) cells, was used to screen for idiotypic antibodies from these hybridomas. The results showed that G_4 hybridoma produced an anti-idiotypic antibody which could bind to the D_3 RGNTF-McAb and neutralize the inhibitory function of D3 on Rb cells, and possessed the trophic activity similar to RGNTF in promoting the growth activity of Rb cells. Immunohistochemical studies with anti-idiotypic antibody revealed that Rb cells and embryonic and neonatal rat retinal ganglion cells possessed coarse immunoreactive positive granules distributed on their cell surface membrane. These granules are identified as the receptors for RGNTF. Therefore, we concluded that the G_4 monoclonal antibody is not only an anti-idiotypic antibody but also an anti-paratopic antibody, It may be very useful in studying the mechanism of action of RGNTF and has a widespread applications in future.

本实验用Phast system电泳法,从大鼠视网膜神经节细胞诱向因子(retinal ganglion neuronotrophic factor,RGNTF)单克隆抗体(RGNTF-McAb)中,分离出一种免疫球蛋白lgG,免疫Balb/c小鼠,制备了抗RGNTF-McAb的独特型单克隆抗体。用MTT微量比色法,对该独特型单克隆抗体进行筛选。结果表明,独特型单克隆抗体G_4能特异性地和RGNTF单克隆抗体D_3结合,中和D_3对成视网膜细胞瘤(Rb)细胞的抑制作用;具有RGNTF的生物学效应,能促进Rb细胞的生长。用免疫组织化学方法证明,Rb细胞、大鼠胚胎视网膜节细胞、新生大鼠的视网膜节细胞都可对G_4独特型抗体发生阳性免疫反应,阳性颗粒较粗,主要分布在Rb细胞和视网膜节细胞的胞膜上,这些阳性颗粒可能是RGNTF的膜受体。所以我们认为,G_4单克隆抗体不仅是独特型抗体,而且是对位型的独特型抗体。它的产生对深入研究RGNTF的作用机制具有重要意义,亦有广阔的应用前景。

MTT colorimetric microassay was used to quantify the effect of various concentrations of tectal extracts (Te) from neonatal rats of defferent age (postnatal day 0-12) on the survival growth of retinal cells. It was found that the survival and growth enhancing effect of Te on the retinal cells increased with increasing Te concentration with a maximal effect achieved at 15 ul/well (about 40 ng protein). The survival and growth enhancing effect of Te from postnatal day 0-2 on the retinal cells was maximum. With further development there was a decline in the survival and growth enhancing effect of the Te. By postnatal day 12, the survival and growth enhancing effect of Te on the neonatal rat retinal cells had declined to a minimum with no difference from cultures without Te (p>0,05). The results suggest that the neuronotrophic effect of Te is changing With neonatal age.

Results showed that both Rb cells were stained immunoreactively strong-positive. Extract prepared from Rb Y-79 cells was run Western blot on a PVDF membrane and only a single immunor-eactively positive protein band was stained at a 30 kD position by the RGNTF-McAb while the rest of protein hands were negative. In order to identify whether both RGNTFs from the Rb cells and tectum having a similar biological trophic activity, horseradish peroxidase-retrograde-prelabelled-retinal ganglion cells (HRP-RGC) from neonatal rats were allowed to culture for 48 h on a native Phastgel with proteins from the Rb extract (Rbe) and tectal. extract (Te) electrophoretically separated. Results showed that HRP-RGC were survived and grown only on the 30 kD band of the Rbe and Te and indicated that the RGNTFs from Rbe and Te have a similar biological trophic activity to RGC. Further evidence suggested that the excessive growth of Rb tumor may be related to its autocrine function of RGNTF.

用抗大鼠视网膜神经节细胞诱向因子(Retinal ganglion neuronotrophic factor,RGNTF)单克隆抗体对临床手术切除的4例人视网膜母细胞瘤(Rb)和培养的人(Y-79)视网膜瘤细胞株进行了免疫细胞化学染色。结果显示,瘤细胞有较强的反应阳性。为进一步证实,用Y-79细胞株制备Rb细胞提取液作快速电泳,并转移至PVDF膜,用RGNTF单克隆抗体进行Western印迹反应,结果表明,RGNTF单抗仅在Rbe电泳的30kD蛋白带呈现单一的阳性反应带,其余蛋白带皆为阴性。为识别Rb瘤细胞内存在的尽GNTF与大鼠顶盖提取液中的RGNTF是否享有相同的生物效应,用辣根过氧化物酶(HRP)逆行标记的初生大鼠视网膜神经节细胞(RGC)培养在Rb提取液(Rbe)和顶盖提取液(Te)电泳后的凝胶片上48小时。结果发现HRP阳性的视网膜神经节细胞只存活在Rbe(共有18条蛋白带)与Te(共有25条蛋白带)中的30kD蛋白带上,表明Rbe中的RGNTF对RGC有同样生物效应。以上结果说明,人视网膜母细胞瘤内存在与大鼠相同的RGNTF,并提示Rb肿瘤细胞的超常生长可能与其自身产生的RGNTF有关。
