Presence of estrogen receptors (ER) and progesterone receptors (PR) in breast cancer tissue have been shown to be useful in predicting those patients who will respond to hormone therapy and have a good prognosis. However, ia some cases, the results of receptor assay do not correspond well to hormone therapy. Although there have already been several possible explanations, we believe a pathological check on the tumor tissuebeing assayed is very important. To test this belief, 684 specimens from 5 hospitals were analyzed for both tumor cell content and steroid receptor content. Our results indicat that there is a direct relationship between percent tumor present and mean receptor con tent. Statistic analysis shows the results are significant. Therefore,good tumor specimens are needed for accurate assays. Secondly, those patients whose tumors contain low amount of tumor cells may result in a false negative ER assay, but due to the larger variation in the PR levels, the tumor will still be classified as PR( + ) in some patients. This might partly explain the unusual ER (-) /PR( + ) status.

The results showed that a detectable rate of ER positive is 59.52% by the single point assay and is 45.24% by a multiple point assay. No significant difference between these results was found by x2 test (P>0.1). For the single point assay, 16 ER positive and 14 ER negative accorded with the results by the multiple point assay. The total percentage in coincidence with ER positive and ER negative of the multiple point assay is 71.4%. An average binding capacity of ER is 38.22±6.18 fmol/mg cytosol protein using the single point method, and is 27.91 ± 5.28 fmol/mg protein using by the multiple point method. Both of them have no significant difference (P>0.3). A quantitative correlation of receptor level between the 2 assays is very remarkable (r=0.6981 P<0.001). We consider that the single point assay might be used clinically after obtaining experience which was assayed by the single point assay as compared to the multiple point assay in given numbers of human breast cancer tissue.

Six hybridoma cell lines against human mammary tumor glycoproteins extract were raised in our laboratory. One of them (GP1D8) was investigated.McAb GP1D8 can react with 82% human breast cancer tissue sections when using the indirect immunoperoxidase histochemistry assay. It has weak reaction with human thyriod carcinoma and normal colon tissue section, and has negative reaction with normal mammary gland and other normal tissues or tumors.The McAb can recognize the 66kd band in SDS-PAGE of the extract....

Six hybridoma cell lines against human mammary tumor glycoproteins extract were raised in our laboratory. One of them (GP1D8) was investigated.McAb GP1D8 can react with 82% human breast cancer tissue sections when using the indirect immunoperoxidase histochemistry assay. It has weak reaction with human thyriod carcinoma and normal colon tissue section, and has negative reaction with normal mammary gland and other normal tissues or tumors.The McAb can recognize the 66kd band in SDS-PAGE of the extract. The value of McAb GPlD8's affinity content is 3.2×10-9M-1 by using competative ELISA. The extract of cytosol glycoproteins has five peaks when using Sepharcryl S-200 HR chro-matography, and the antigen activity is in the second peak. Other experiments show that antigen is a polypeptide with about 16% carbohydrate. It can remain its antigen activity after 30min of 70℃ treating. The practical use of McAb GP1D8 was under investigation.

以人乳腺癌组织提取的糖蛋白免疫后的BALB/C小鼠的脾细胞与小鼠骨髓瘤细胞系SP2/0细胞融合,经筛选及克隆化培养,建立了1株稳定分泌特异性抗体的杂交瘤细胞株GP-1D_8。 McAb GP-1D_8与乳腺癌组织产生强阳性反应;与正常乳腺及一些其它肿瘤组织为阴性反应;与甲状腺癌及正常结肠组织出现弱交叉反应。 经Sepharcyl-S200 HR分子筛分离的抗原分子,分子量为66kd,与Western blot测出的分子量相符。抗原分子含糖量为16%。其抗原决定簇为一段耐70℃温度的多肽。与前人报导的乳癌相关抗原有区别,可能是一种新的乳腺癌相关抗原。
