The results showed that: (1) The proliferation activity of splenic lymphocytes stimulated by Con A was significantly increased with microinjection of Leu-enkephalin 1μl (18 mmol/L) or Met-enkephalin 1 μl (1 8 mmol/L) into bilateral hippocampus, but was inhibited by Lipopolysaccharide 1μl (50ng/μl). The proliferative response of splenic lymphocytes to Leu-enkephalin still persisted after bilateral adrenalectomy. Low doses of Leu-or Met-enkephalin (10-10, 1 0-11 mol/L) were also capable of causing proliferative activity of the Con A- stimulated cultured splenic lymphocytes in vitro. (2) Using RT-PCR technique, IL- 1α gene expression was detected in hippocampus 90 min after bilateral intrahippocampal injection of LPS. The LPS-induced IL-1α gene expression in hippocampus could ilo longer be detected 30 min aftter microinjection of Met-or Leu-enkephalin into hippocampus. The above results suggested that intrahippocampal enkephalin might play an important role in neuro-immunomodulation by enhancing the inhibition of IL-1α gene expression in hippocampal formation.

本文研究了海马内微量注射脑啡肽对大鼠细胞免疫功能及海马内白细胞介素-1α(IL-1α)基因表达的影响。结果发现:(1)双侧海马内微量注射甲硫-脑啡肽或亮-脑啡肽各1μl(18mmol/L)能明显增强ConA刺激的脾淋巴细胞增殖活性,而双侧海马内微注射脂多糖各1μl(50ng/μl)则起抑制作用。切除双侧肾上腺后,海马内注射脑啡肽仍可增强脾淋巴细胞增殖反应。低浓度亮-及甲硫-脑啡肽(10 ̄(-10),10 ̄(-11)mol/L)直接作用于体外培养的大鼠脾淋巴细胞,也明显增强ConA刺激的细胞增殖活性。(2)双侧海马内微量注射脂多糖各1μl(50ng/μl),于90min后用逆转录-聚合酶链反应技术检测到海马结构内IL-1α基因表达,而海马内甲硫或亮脑啡肽注射30min后再给予脂多糖,则未检测到IL-1αcDNA的扩增条带。上述结果表明,海马结构对机体免疫功能的调节中脑啡肽起着重要作用,其机制可能与抑制海马结构IL-1α基因表达有关。

The P300 potentials in the skull and the hippocampus were recorded synchronically by implanting electrode into bilateral hippocampus of rabbits by means of stereotaxic technic P300 recorded on the hippocampus showed a polarity inversion and its amplitude was obviously increased,especially P3b.However,P300 recorded on the skull turned into a single spike,P3b disappeared and P3a latency prolonged,suggesting that P3b was originated from the hippocampus.

Objective:To investigate the effects of ciliary neurotrophic factor(CNTF) on neural damage in hippocampal CA3 induced by stress. Methods: The changes of morphology of hippocampal neurons during acute or chronic footshock stress in rats, and the effects of CNTF administrated into bilateral hippocampus on the changes were observed by Nissl staining, Bielschowsky Gros Lawrentjew staining and electron microscopy. Results: The morphology of CA3 neurons in the hippocampus was not changed during acute stress in rats but changed during chronic stress. CNTF administrated into bilateral hippocampus did not effect the morphology of CA3 neurons in the control and acute stressful rats, but significantly reduced CA3 neuronal damage induced by chronic stress. Conclusion: CNTF can protect CA3 neurons from damage induced by chronic stress.

目的 :探讨睫状神经营养因子对应激引起海马 CA3区神经元损害的作用。方法 :采用 Bielschowsky- Gros-Lawrentjew染色法和常规透射电镜技术 ,观察急性和慢性足底电击大鼠的海马 CA3区神经元形态的变化 ,及双侧海马注射睫状神经营养因子对其影响。结果 :急性应激大鼠海马神经元形态无明显变化 :慢性应激大鼠海马神经元出现明显的损伤性形态变化 ;睫状神经营养因子对正常大鼠和急性应激大鼠的海马神经元形态无明显作用 ,但可显著减轻海马神经元损伤程度。结论 :睫状神经营养因子可能通过保护海马 CA3区神经元从而改善慢性应激大鼠的行为障碍
